摘要： Antisense oligonucleotide gene therapy selective for the 5' region of PDGFR-.bet a. subunit mRNA was used in attempt to prevent intimal thickening following rat car otid arterial injury. Sustained perivascular application of the antisense oligomers f or 14 days reduced PDGFR-.beta. protein overexpression and prevented neointima formati on by 80%. Specificity was verified by the absence of effects on the expression of a non-targeted gene PDGFR-.alpha.. These data demonstrated that antisense oligonuc leotide sequences can effectively suppress a growth factor receptor, and the reduction o f intimal hyperplasia after injury correlates with the extent to which these oligo mers inhibited PDGFR-.beta. protein expression. Methods and materials useful for prev enting restenosis are described and claimed.

摘要： Methods for treating or preventing neointima stenosis are disclosed. The methods generally involve the use of a TGFbeta inhibitor, a SMAD2 inhibitor, an FGF Receptor agonist, a Let-7 agonist, or a combination thereof, to inhibit endothelial-to-mesenchymal transition (Endo-MT) of vascular endothelial cells into smooth muscle cells (SMC) at sites of endothelial damage. The disclosed methods can therefore be used to prevent or inhibit neointimal stenosis or restenosis, e.g., after angioplasty, vascular graft, or stent. Also disclosed are methods for increasing the patency of biodegradable, synthetic vascular grafts using a composition that inhibits Endo-MT. A cell-free tissue engineered vascular graft (TEVG) produced by this method is also disclosed.
