The immature DCs generated and maintained in this manner were CD14 and expressed high levels of CD1a. Upon maturation by contact with an agent such as, for example, BCG and IFNgamma, the cells were determined to express surface molecules typical of mature dendritic cells purified by prior methods and cultured in the presence of GM-CSF and IL-4. The mature dendritic cells produced from monoxytes without activation and cultured in GM-CSF alone are suitable for use in dendritic cell-based immunotherapy methods, such as for use in the treatment of disease, including cancer.

Al evitar la activacion, tal como por ejemplo al evitar la union de las celulas a la superficie del recipiente de cultivo, los monocitos no requieren la presencia de citosinas adicionales, tales como por ejemplo IL-4 o IL-13, para evitar la diferenciacion en un linaje de celulas no dendriticas. Las DC inmaduras generadas y mantenidas de esta forma fueron CD14- y expresaron altos niveles de CD1a. En el momento de la maduracion mediante el contacto con un agente tal como por ejemplo, BCG e IFN?, se determino que las celulas expresan moleculas superficiales tipicas de las celulas dendriticas maduras purificadas por los metodos anteriores y los cultivos en presencia de GM-CSF e IL-4. las celulas dendriticas maduras producidas a partir de monocitos sin activacion y cultivadas en el GM-CSF solo son adecuadas para utilizarse en celulas dendriticas con base en metodos de monoterapia, tales como por ejemplo para utilizarse en el tratamiento de enfermedades, entre las que se incluyen el cancer.
