摘要： PROBLEM TO BE SOLVED: To efficiently screen and breed the subject new yeast with high bread dough fermentative power, capable of producing bread with good flavor, in particular good smell by using acetoin as an indicator. SOLUTION: This new yeast having high bread dough fermentative power and/or being capable of producing bread with good flavor, in particular good smell is obtained by the following process: a strain capable of highly producing acetoin is screened from a cultured product afforded through pref. aerobic culture; in particular, such a strain as to be >=600 ppm (pref. >=800 ppm) in the acetoin content of the supernatant of a cultured product afforded by aerobic culture in a modified schultz-Atkin medium for 48 h is screened; preferably, diploid yeast capable of highly producing acetoin are crossbred to create excellent tetraploid bread yeast.

The use of the yeast makes the fructooligosaccharide added to the raw material remain without decomposition.

SOLUTION: This harvest of a fission yeast polyploid strain comprises inoculating the fission yeast such as Schizosaccharomyces pombe JY1 strain to a medium, shaking the medium at 30 deg.C for 30 hours, centrifuging the culture solution to collect the yeast, washing the yeast with 0.86% NaCl aqueous solution, suspending the yeast in 0.86% NaCl aqueous solution, enclosing the yeast suspension in a polyethylene container, treating the yeast under 100-250Mpa hydrostatic pressure at 25 deg.C for 10 minutes using a pressure treating device, culturing the fission yeast after the heat treatment on an agar medium containing a pigment and selecting the polyploid colonies based on a color change. Thus, a screening can be carried out simply and accurately and the polyploid strain of the objective fission yeast can be effectively obtained in an extremely high frequency.
