摘要： The present invention provides a recombinant soluble trimeric hemagglutinin (rHA) protein comprising a hemagglutinin ectodomain and an oligomerization domain. The rHA is produced as a soluble homotrimer, and may further comprises a signal peptide and/or an endoplasmic reticulum (ER) retention signal. The invention is also directed to nucleic acids encoding the rHA of the invention, as well as vectors and chimeric constructs comprising the nucleic acid. Methods of producing the rHA are also provided. The rHA described herein may be used to formulate influenza vaccines, or may be used to enrich existing vaccines.

摘要： A method for synthesizing a protein of interest within a plant or a portion of a plant is provided. The method involves introducing one or more than one nucleic acid sequence encoding a protein of interest operatively linked with a regulatory region obtained from a photosynthetic gene that is active in the plant, in a transient manner. The plant is then maintained under conditions that permit the nucleic acid sequence encoding the protein of interest to be expressed in the plant or the portion of the plant. The plant may be pruned prior to the introducing one or more than one nucleic acid sequence.

摘要： A nucleic acid encoding an affinity trapping agent is provided. The affinity trapping agent comprises a peptide or protein ligand having a binding affinity for a protein of interest, fused to a heterologous subcellular compartment targeting signal. Methods for targeting the immobilization of a recombinant peptide or protein of interest, or an endogenous peptide or protein of interest, to a subcellular compartment of a cell are also provided. If the peptide or protein of interest is recombinant then a nucleotide sequence encoding the recombinant peptide or protein of interest and a nucleic acid encoding the affinity trapping agent are introduced into the cell and co- expressed to form a complex thereby targeting the immobilization of the recombinant peptide or protein of interest. If the peptide or protein of interest is endogenous, then the nucleic acid encoding the affinity trapping agent having a binding affinity for the endogenous peptide or protein of interest is introduced into the cell, and the nucleic acid encoding the affinity trapping agent is expressed in the cell to form a complex thereby targeting immobilization of the endogenous peptide or protein of interest.

The method involves expression of influenza HA of type A/California/04/09 in plants and the purification by size exclusion chromatography. The invention is also directed to a nucleic acid encoding influenza HA of type A/California/04/09 as well as vectors. The VLPs may be used to formulate influenza vaccines, or may be used to enrich existing vaccines.

摘要： Methods of preparing plant-derived virus like particles (VLPs) are provided. The method may comprise obtaining a plant, or plant matter comprising apoplast-localized VLPs, producing a protoplast/spheroplast fraction and apoplast fraction from the plant or plant matter, and recovering the apoplast fraction. The apoplast fraction comprises plant-derived VLPs. Alternatively, VLPs may be obtained from plant or plant matter comprising plant-derived VLPs by digesting the plant matter using a cell wall degrading enzyme composition to produced a digested fraction. The digested fraction is filtered to produced a filtered fraction, and the plant- derived VLPs are recovered from the filtered fraction.

The method comprises co-expressing within a plant a nucleotide sequence encoding a first nucleotide sequence encoding a hybrid protein (GNTl-GaIT) comprising a CTS domain of N-acetylglucosaminyl transferase (GNTl) fused to a catalytic domain of beta-l,4galactosyltransferase (GaIT), the first nucleotide sequence operatively linked with a first regulatory region that is active in the plant, and a second nucleotide sequence for encoding the protein of interest, the second nucleotide sequence operatively linked with a second regulatory region that is active in the plant. The first and second nucleotide sequences are co-expressed to synthesize a protein of interest comprising glycans with the modified N-glycosylation profile within the plant, the portion of the plant, or the plant cell.

The amount of medicagenic acid saponin in the preparation is greater than 50% by weight to produce the cholesterol-lowering effect in an animal. A method of purifying a preparation of at least 30% medicagenic acid saponin is also disclosed. The preparation may be purified from alfalfa plants and used as a treatment for lowering cholesterol and triglycerides in an animal, in particular a human.

摘要： A method of synthesizing sialic acid in plants, and plants capable of synthesizing sialic acid is provided. Furthermore, a method of producing sialylated protein in a plant is also provided. 摘要： A method of producing a virus like particle (VLP) in a plant comprising modified hemagglutinin is provided. The method comprises introducing a nucleic acid comprising a regulatory region active in the plant and operatively linked to a nucleotide sequence encoding a modified influenza hemagglutinin (HA) protein into the plant, or portion of the plant, the modified HA protein comprises a modified proteolytic loop. Followed by incubating the plant or portion of the plant under conditions that permit the expression of the nucleic acids, thereby producing the VLP. The modified proteolytic loop may comprise one or more protease cleavage sites exhibiting reduced or abolished cleavage by a protease. The nucleotide sequence encoding the HA may be selected from the group consisting of B HA, C, H1, H2, H3, H4, H5, H6, H7, H8, H9, H10, H11, H12, H13, H14, H15, and H16. Also described is a virus like particle (VLP) produced by the method, and plants expressing the VLP. The virus like particle (VLP) may comprise plant-specific N-glycans, or modified N- glycans.
