    • Reaction is effected in a water-immiscible but substrate-miscible organic solvent in order to keep the substrate conc. in the aq. medium lower than that inhibiting the enzyme activity. Process is applicable to reaction of indole with L- or DL-serine into L-tryptophan by tryptophan synthetase produced by E. coli, where the following organic solvent may be employed; aromatic hydrocarbons (e.g. benzene, toluene, chlorobenzene, nitrobenzene, acetophenone), aliphatic esters of 6C or higher (e.g. n-BuOAc, i-AmOAc, EtOCOPr, i-BuOAc), citric acid esters (e.g. acetyltriethyl citrate, acetyltributyl citrate, triethyl citrate, tributyl citrate), tartaric acid esters, malic acid esters, ethers (e.g. anisole). In producing tryptophan, a variant of E. coli is cultured in an aerobic condition at 28-40 deg. C or pH 6-8, and the culture broth or the cells sepd. from the broth are dispersed into a soln. contg. pyridoxal HCl, L-serine and inorganic materials. A soln. of indole dissolved in the above solvent (pH 7.5-9.5 (8-9)) is added in one portion or successively in small portions to the above prepd. suspension and kept at 20-40 deg. C to yield L-tryptophan. The indole content in the solvent may be fixed to be 800 (750 ppm or lower) in industrial scale prodn. In certain reactions utilising enzymes, excess content of substrates sometimes deteriorates enzyme activity to decrease the yield of desired prod. Deterioration is now avoided by controlling substrate concn. in the reaction medium.
