摘要： PROBLEM TO BE SOLVED: To obtain a new gene coding for chondromodulin II protein having a specific molecular weight, having osteoclast growth activity, chondrocyte differentiation function promoting activity and chondrocyte growth activity and capable of giving a protein useful for treating bone or cartilage diseases. SOLUTION: This new DNA codes for chondromodulin II protein, which has an amino acid sequence at least from 1 to 133 of amino acid numbers in the amino acid sequence expressed by the formula, etc., is composed of a water-soluble protein consisting of a kind of polypeptide, has about 16 KD in a molecular weight measured by SDS-polyacrylamide electrophoresis, and has activity for growing osteoclasts and chondrocytes by themselves or in the presence of another cell growth factor and activity for promoting differentiation function against chondrocytes. The DNA is useful as an active component of a medicinal composition for treating bone or cartilage diseases. The DNA can be obtained by cloning a cDNA library prepared from bovine cartilage through PCR using a primer having a partial base sequence.

A transformant transformed with the resultant expression vector is then cultured to thereby produce the human chondromoduin-I protein.

摘要： PURPOSE:To obtain a new chondromodulin-I protein having a specific molecular weight, growth activities of a chondrocyte, promoting activities of the differentiation function thereof and suppressing activities in growing a vascular endothelial cell and useful for treating fracture, various chondropathies, cancer., etc. CONSTITUTION:This new human chondromodulin-I protein is obtained by screening a genomic library derived from a human using a probe composed of a part of a gene coding for a bovine chondromodulin, sorting a positive clone, recovering a DNA from the resultant clone, treating the DNA with a restriction enzyme, then joining the prepared DNA fragment to a suitable vector, providing an expression vector containing the DNA of coding for a human chondromodulin, then transducing the obtained expression vector into a host such as an animal cell, transforming the host and subsequently culturing the resultant transformant. The prepared human chondromodulin-I protein has about 26 kdaltons molecular weight measured by an SDS-polyacrylamide gel electrophoresis and is capable of growing a chondrocyte alone or in the coexistence of a fibroblast growth factor, promoting the differentiation function of the chondrocyte and suppressing the growth of a vascular endothelial cell.

摘要： PROBLEM TO BE SOLVED: To obtain a promoter of bone resorption having bone resorbing activities, effective to metabolic bone disease by including angiogenin as an active ingredient. SOLUTION: This promoter of bone resorption includes angiogenin having an amino acid sequence of the N-terminal of a protein of the formula as an active ingredient. The angiogenin is preferably purified by performing ultrafiltration, gel-filtration, heparin affinity chromatography, reversed-phase HPLC method of the extract of bovine fetus cartilage. Further, a part of amino acids of the angiogenin can be subjected to variation such as substitution, modification or addition within the range not weakening the bone resorbing activities.

摘要： PURPOSE:To obtain an osteoclast formation and bone resorption promoter effective in treating metabolic osteopathy for which there has not been any effective therapy up to the present. CONSTITUTION:This osteoclast formation and bone resorption promoter contain a cartilage growth factor having the following physico-chemical properties as an active ingredient: (1) about 16000 Da molecular weight measured by a sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis; (2) having activities in proliferating a cartilage alone or in the coexistence of a fibroblast growth factor and (3) having activities in promoting the differentiating functions of the cartilage. The cartilage growth factor preferably has an N-terminal amino acid sequence represented by formula I and a partial amino acid sequence represented by formulas II to IV. A substance having inhibiting actions on the bone resorption of the medicine is regarded as capable of suppressing the formation of an osteoclast and having effects on suppression of the bone resorption. Thereby, a suitable evaluation system is used to screen a substance capable of inhibiting biological activities of the cartilage growth factor. As a result, the medicine for osteopathy such as osteoporosis is simply obtained.
