摘要： The use of a compound of formula I or a pharmaceutically acceptable salt thereof or a prodrug thereof in the manufacture of a medicament for the treatment and/or prevention of hearing loss including age-related hearing impairment, and tinnitus: wherein Y represents hydrogen or C 1-6 alkyl; Z represents C 1-6 alkyl, C 3-7 cycloalkyl, C 4-7 7 cycloalkenyl, aryl, C 3-7 heterocycloalkyl, heteroaryl or di(C 1-6 )alkylamino, any of which groups may be optionally substituted; or Y and Z are taken together with the two intervening carbon atoms to form a ring selected from C 5-9 cycloalkenyl, C 6-10 bicycloalkenyl, tetrabydropyridinyl, pyridinyl and phenyl, any of which rings may be optionally benzo-fused and/or substituted; R1 represents C 3-7 cycloalkyl, phenyl, furyl, thienyl or pyridinyl, any of which groups may be optionally substituted; and R2 represents cyano(C 1-6 )alkyl, hydroxy(C 1-6 )alkyl, C 3-7 cycloalkyl(C 1-6 )alkyl, propargyl, C 3-7 heterocycloalkylcarbonyl(C 1-6 )alkyl, aryl(C 1-6 )alkyl or heteroaryl(C 1-6 )alkyl, any of which groups may be optionally substituted. These compounds are selective ligands for GABA A receptors, in particular, having high affinity for the a 2 and/or a 3 subunits thereof. The compounds may be formulated in a variety of solid or liquid forms for internal application by various routes such as oral, intranasal or parenteral.
