    • The invention relates to an anti-tumour antibiotic, Iyomycin complex, containing an acidic polypeptide as a major component A and a basic substance as a minor component B, and having the following properties: ultra-violet absorption spectrum showing a (Fig. 1, not shown); infrared spectrum having absorption maxima at 2.95 m and 6.05 m (Fig. 2); elementary analysis C = 47.64%, H = 6.15%, N = 9.4%, ash 8.3%, total N 8.8-9.1%; positive reactions to ninhydrin, xanthoprotein, Folin and Pauli reagents and negative to Millon, Molisch, Tollens, Benedict, Elson-Morgan, biuret, Adamkiewitz, Liebermann, Sakaguchi and nitroprusside reagents; on acid hydrolysis produces glutamic acid, aspartic acid, serine, threonine, proline, glycine, alanine, valine, leucine, phenylalanine, tyrosine, cysteine and lysine. Iyomycin is produced by cultivating Streptomyces phaeoverticillatus nov. spec., especially NRRL 3021, in an aqueous nutrient medium containing a source of assimilable carbon, nitrogen and inorganic salts, e.g. aqueous solution containing meat extract, sodium chloride, calcium carbonate, soyabean oil, peptone, and glucose. Iyomycin complex is obtained from the clarified broth by (A) treatment with 1% magnesium sulphate or passage through cation exchange resin in sodium form to remove impurities, (B) precipitation by adjusting to pH 3.6 (isoelectric point) or adding a salting-out agent such as ammonium sulphate, magnesium sulphate, a salt of a heavy metal, e.g. zinc chloride, lead acetate, cupric sulphate or ferrous sulphate, aluminium hydroxide, or 6,9-diamino-2-ethoxyacridine lactate, (C) treating precipitate (B) with water, dilute alkali or phosphate buffer and removing inorganic ions by dialysis. Further purification is by passage through column of "Sephadex G25" (Registered Trade Mark).
