摘要： PROBLEM TO BE SOLVED: To provide a method for separating and collecting mesenchymal stem cells to utilize them for the regenerative treatment of a tissue, etc., and for the preparation of mammalian cells used for the regeneration and/or restoration of the tissue, safe for a collected parent body and also easy for collection, and also a practical method for preparing cells for transplant by using the collected mesenchymal mast cells. SOLUTION: This method for collecting mesenchymal stem cells capable of collecting the cells by using a method for separating and collecting the cells from an oral cavity tissue by a simple operation of requiring a minimum exfoliation and incision of the skin and muscle, not imposing a burden to the collected parent body, safely and simply collecting the necessary amount of the cells maintaining an excellent differentiation potency. Further, the use of the mesenchymal stem cells is provided by culturing and differentiation-inducing the separated and collected mesenchymal stem cells to prepare cells used for the regeneration and/or restoration of a tissue having high differentiation potency, and transplanting the prepared cells to the defective part and/or part for restoration such as the bone, cartilage, periodontal tissue, etc., to regenerate and/or restore the tissues.

摘要： PROBLEM TO BE SOLVED: To provide a new method for culturing a mesenchymal stem cell capable of obtaining extremely larger amount of the mesenchymal stem cell than a conventional method under practical conditions aiming the use of the mesenchymal stem cell for a regenerative care of a tissue. SOLUTION: It was found that, by culturing a mesenchymal stem cell in the presence of an extracellular substrate of basement membrane cells, for example, by culturing a mesenchymal stem cell on a culture dish coated with the extracellular substrate of basement membrane cells, the mesenchymal stem cell grows at an extremely high speed, it can keep a differential ability to an osteoblast, a chondrocyte or a fat cell, it can be effectively grown even with a serum of a low concentration, it can be cultured by using human serum, and further, excellent differentiation-inducing culturing is possible. Further, even in the case where culturing is carried out in a lower concentration of serum, growth ability and differential ability can be kept by adding various kinds of additives to the culture medium.

摘要： PROBLEM TO BE SOLVED: To provide a method for effectively identifying and isolating a mesenchymal stem cell and a fibroblast the distinction of which is difficult because of resembled shapes by using a gene marker and/or a protein marker. SOLUTION: The mesenchymal stem cell and the fibroblast are identified/isolated by using a gene of a vitamin D receptor or a protein thereof, or a vitamin D receptor-associated gene such as osteocalcin and osteopontin, or a protein thereof as a fraction marker of the mesenchymal stem cell and the fibroblast, and detecting the gene and the expression of the protein in a subject cell. The identification/isolation of the mesenchymal stem cell is effectively carried out by detecting the expression of an activated gene because the expression of the target gene of the vitamin D receptor is activated in the mesenchymal stem cell by the addition of a material for activating the expression of the gene such as 1,25-dihydroxyvitamine D.
