摘要： Provided is a biomimetic ice-inhibiting material. A library of structures of compound molecules is constructed, wherein the compound molecule comprises a hydrophilic group and an ice-philic group. Next, spreading performance of each compound molecule at an ice-water interface is evaluated by means of molecular dynamics simulation (MD simulation). Finally, a compound molecule having the desired ice affinity and water affinity is selected by means of screening. The present application proposes for the first time a mechanism of ice-philicity and hydrophilicity of an ice-inhibiting material, applies MD simulation for the molecular structure design of the ice-inhibiting material, evaluates the ice-philicity and the hydrophilicity of the designed ice-inhibiting material by means of MD simulation, predicts ice inhibition performance of the ice-inhibiting material, and optimizes the structure thereof. The biomimetic ice-inhibiting material designed and selected by the method greatly inhibits the growth of ice crystals. When added to a cryopreservation reagent, the biomimetic ice-inhibiting material achieves a good cryopreservation effect even without DMSO.

摘要： Disclosed by the present invention is a cryopreservation solution without DMSO. On the basis of a volume of every 100 mL, the cryopreservation solution contains 0.01-50.0 g of bionic ice control materials, 5.0-30 mL of polyols, 1-30 g of water-soluble sugar, and 0-30 mL of serum, the rest being a buffer solution. The bionic ice control materials are selected from PVA and/or amino acid based bionic ice control materials. The cryopreservation solution according to the present invention and a freezing balancing solution do not contain DMSO. When being used for the cryopreservation of mouse oocytes and embryos, the described solution may achieve the same or an even higher cell and tissue survival rate and functional expression stability as or than a commercial cryopreservation solution (containing 15% DMSO), and has high preservation efficiency. The cryopreservation solution without DMSO or serum further solves the problems wherein parasitic biological contaminants may be introduced and stability is poor due to the commercial cryopreservation solution currently commonly used in clinical practice containing serum. The cryopreservation solution according to the present invention has a simple composition, a convenient source of raw materials and is low-cost, and may be widely applied in the cryopreservation of various types of cells.

摘要： The present invention relates to a biomimetic ice growth inhibition material, which is prepared by constructing a library for structures of compound molecules, with the compound molecules comprising a hydrophilic group and an ice-philic group, by evaluating the spreading performance of each compound molecule at an ice-water interface by adopting molecular dynamics simulation (MD simulation), and by screening the compound molecules with the desired affinities for ice and water. The present invention firstly provides the mechanism of the affinities of the ice growth inhibition material for ice and water, introduces MD simulation into the molecular structure design of the ice growth inhibition material, evaluates the affinities of the designed ice growth inhibition material for ice and water through MD simulation, predicts the ice growth inhibition performance of the ice growth inhibition material, and can realize the optimization of the structure. The biomimetic ice growth inhibition material designed and screened by the method disclosed herein has good capability of inhibiting the growth of ice crystals, and can achieve a good cryopreservation effect on reagents even without adding DMSO.

摘要： Disclosed in the present invention are a thawing solution for cryopreservation, a preparation method therefor and a use thereof. The thawing solution comprises: 0.1-50 g of a biomimetic material for controlling ice, 0-1.0 mol L -1 of a water-soluble sugar, and balance of a buffer solution, wherein the biomimetic materials for controlling ice is a material for controlling ice having an ice-philic group and a hydrophilic group, the hydrophilic group is a functional group that can form a non-covalent interaction with water molecules, and the ice-philic group is a functional group that can form a non-covalent interaction with ice. In the present invention, by means of the thawing solution and the thawing reagent prepared from the biomimetic material for controlling ice, which material has ice-philic and hydrophilic properties, the growth and recrystallization of ice crystals can be effectively controlled, and damage to cells or tissue caused by uncontrolled growth of the ice crystals during recovery can be significantly ameliorated. Moreover, the thawing solution and the thawing reagent have a good biocompatibility, do not contain animal serum, and have a low toxicity. Compared with a traditional thawing solution containing serum, the thawing solution and the thawing reagent of the present invention reduce the risks of shelf life being short and introduction of parasitic biological contaminants, and are more conducive to maintaining the generation stability of cells or tissues. In addition, the present invention has simple components, is low in cost and has good application prospects.
