    • PROBLEM TO BE SOLVED: To provide a bisulfite reaction which can be readily performed manually, a bisulfite reaction using a solid phase, particularly magnetic glass particles, which is handleable by routine analytical equipment, and a bisulfite reaction performed successfully in a satisfying manner since the nucleic acid is bound to the surface of the solid phase by various interactions possibly influencing the success of the bisulfite reaction. SOLUTION: A method for the conversion of cytosine bases in a nucleic acid to uracil bases, wherein the nucleic acid is bound to a solid phase during a deamination step and/or a desulfonation step, the use of the solid phase in the nucleic acid during the deamination step and/or desulfonation step of the reaction in which cytosine bases in the nucleic acid are converted to uracil bases in the presence of bisulfite ion, and a kit for performing bisulfite reaction, containing a solution containing the bisulfite ion and a solid phase are provided.
