    • A method of providing a document which can be visually authenticated includes providing a document that is valid for a predetermined period of validity; randomly selecting visual authentication indicia just prior to the period of validity, the visual authentication indicia providing visual indication of validity during the period of validity; and transferring the visual authentication indicia to the document to visually authenticate the validity of the document during the period of validity. The method can be implemented using a system for creating a document which can be visually authenticated disclosed herein. The system includes a plurality of possible visual authentication indicia; a computer capable of executing instructions for randomly selecting one of the plurality of possible visual authentication indicia just prior to a predetermined period of validity; and at least one document validation station for transferring the selected visual authentication indicia to a document to visually authenticate the validity of the document during the period of validity.

    • The present invention provides a method of producing a cloned insert that is representative of the ends of a large segment of DNA from the genome of an organism. Specifically, the present invention provides a clone insert strategy that comprising the steps of: a) isolating a nucleic acid molecule, or portion thereof, wherein said nucleic acid molecule is at least 20 kb; b) ligating said nucleic acid molecule into a vector molecule; c) cutting said nucleic acid molecule with a restriction endonuclease, wherein said restriction endonuclease cuts said isolated nucleic acid molecule of step (a) in at least two sites but does not cut said vector DNA, d) ligating said cut nucleic acid molecule; e) transforming a host cell with said ligated nucleic acid molecule to propagate said molecule; f) determining the sequence of the ends of the nucleic acid insert and the sequence from the restriction endonculease site.

    • A time indicator rapidly changes color after a specific time interval. The indicator comprises “back part” that includes a base substrate with a colored dye or colorant deposited on a first surface. A colorant impermeable barrier layer overlays the colorant or colored dye. The indicator further comprises a “front part” that includes a substrate having an adhesive on a first surface thereof. When the back part and front part are put into adhesive contact with each other, the parts coact with each other to cause the colorant impermeable barrier layer to change to a colorant permeable layer to permit the dye to migrate through the layers to cause a color change visible through the front part.

    • An indicator is disclosed that detects the presence of a chemical residue. The indicator includes a substrate having an upper surface and a lower surface and a first portion and a second portion joined at a fold line. The first portion is smaller in area than the second portion. An adhesive coats the upper surface of at least the second portion of the substrate. A first reactant is adhered to the upper surface of the first portion of the substrate. When the second portion is contacted with a surface containing the chemical residue, the residue adheres to the adhesive and when the first portion is subsequently folded along the fold line so that the upper surfaces of the substrate contact each other, a portion of the adhesive on the second portion remains exposed and the first reactant and chemical residue react to provide the color indicia indicating the presence of the chemical residue.

    • A time indicator that provides a color indicia after a predetermined period of time has passed after activation. The time indicator includes a substrate, e.g., a clear or transparent substrate, having an upper surface and a lower surface and a first portion and a second portion joined at a fold line. The first portion is of a smaller area than the second portion. An adhesive coats the upper surface of at least the second portion of the substrate. A first reactant is adhered to the upper surface of the first portion of the substrate and a second reactant is adhered to the upper surface of the second portion of the substrate. When the first portion is folded along the fold line so that the upper surfaces of the substrate contact each other, a portion of the adhesive on the second portion remains exposed. This exposed area may be used to attach the indicator to an article or documents. When so folded the first reactant and second reactant contact each other to activate the indicator and to provide the color indicia after the predetermined period of time after activation passes. In one embodiment, the indicator is used to detect the presence of a chemical residue, e.g., explosive compound residues.

    • The invention provides methods and compositions for assembling a modular replacement genome in a host microorganism. After such assembly, the host organism's genome is inactivated or ablated to permit full control of host cellular functions by the replacement genome. A modular replacement genome comprises an assembly of nucleic acid fragments, or segments, derived from one or more natural organisms or from synthetic polynucleotides or from a combination of both. Such an assembly, or set, of segments making up a replacement genome comprises a substantially complete set of genes and regulatory elements for carrying out minimal life functions under predefined culture conditions. The invention provides modular genomes having modules that are amenable to facile replacement, deletion, and/or additions. Such modules may be synthetic polynucleotides and may be designed for controlling gene content, excluding of genes that encode inhibitors or otherwise undesirable competing enzymes that divert a host cell from desired metabolic/synthetic processes; modifying codon usage to maximize or minimize protein production; modifying regulatory elements, including promoters, enhancers, repressors, activator, or the like, to modulate gene expression; balancing enzymatic and transport activities to optimize fluxes of substrates, intermediates, and products in metabolic pathways, and like objectives.
