摘要： The present invention pertains to methods of preventing and eliminating trisulfide bonds in proteins such as antibodies. In one embodiment, trisulfide bonds in proteins are converted to disulfide bonds as part of chromatographic purification procedures. In another embodiment, the formation of trisulfide bonds in proteins is inhibited by implementation of methods described herein during the cell culture production of such proteins. In another embodiment, monoclonal antibodies are produced by the methods described herein.

摘要： The present disclosure is directed to methods to improve the yields of proteins recombinantly produced in eukaryotic cells comprising the addition of anionic substances such as valproate, malate, succinate, fumarate, citrate (e.g., ferric citrate), polyanionic compounds (e.g., polysulfated compounds such dextran sulfate or polyvinyl sulfate), and combinations thereof, wherein the addition of such compounds reduces the binding of recombinant proteins to the cell surface, therefore increasing the amount of soluble protein that can recovered from the culture medium. In addition, the protein yields can be improved according to the disclosed methods by preventing or reducing cell growth inhibition caused by the binding of the expressed recombinant proteins (e.g., neublastin, or antibodies or antigen-binding fragments thereof) to the cell surface. The disclosed anionic substances can also be used during the harvesting phase of a protein production process to release the expressed recombinant proteins from the cell surface prior to protein purification.

摘要： Some embodiments are directed to a process for forming products from animal waste (e.g., poultry litter). The products may include a controlled release fertilizer and renewable natural gas. The process may include providing a feedstock comprising greater than about 80 percent poultry waste and a moisture content of about 25 percent. The feedstock may be diluted to form a slurry having a moisture content of about 90%. The slurry may be mechanically refined to reduce the particle size distribution and the average particle size of the slurry. The slurry may be anaerobically digested to produce biogas and a digestate. The biogas may be converted to renewable natural gas, and the digestate may be converted to a controlled release fertilizer. Additionally, some embodiments are directed to methods of reducing nitrogen emission from soil amendments. Some embodiments are directed to reducing nitrogen emissions sufficient to produce emissions offset credits.

摘要： The present invention is directed to cellulytic host cells. The host cells of the invention expressing heterologous cellulases and are able to produce ethanol from cellulose. According to the invention, host cells expressing a combination of heterologous cellulases can be used to produce ethanol from cellulose. In addition, multiple host cells expressing different heterlogous cellulases can be co-cultured together and used to produce ethanol from cellulose. Furthemore, the invention demonstrates for the first time the ability of Kluveryomyces to produce ethanol from cellulose. The yeast strains and co-cultures of yeast strains of the invention can be used to produce ethanol on their own, or can also be used in combination with externally added cellulases to increase the efficiency of saccharification and fermentation processes.
