    • PURPOSE: A method for modifying DDR2(discoidin domain receptor) tyrosine kinase activity and DDR2 protein having modified kinase activity thereby are provided, thereby treating diseases caused by oxidative stresses and over-activation of DDR2 tyrosine kinase. CONSTITUTION: A method for modifying DDR2(discoidin domain receptor) tyrosine kinase activity comprises the steps of: amplifying cDNA encoding the amino acid sequence containing the DDR2 kinase active site and connecting the amplified sequence to the C-terminal coding region of glutathione-S-transferase(GST) to produce DDR2 kinase-GST fusion gene; cloning the DDR2 kinase-GST fusion gene into an expression vector, expressing the tyrosine kinase active site of DDR2 protein-GST fusion protein in a host cell, and purifying the protein; and adding oxidative stresses to the fusion protein to phosphorylate tyrosine in the tyrosine kinase active site of DDR protein.
    • 目的:提供一种改变DDR2(盘状结构域受体)酪氨酸激酶活性的方法,从而提供具有修饰的激酶活性的DDR2蛋白,从而治疗由氧化应激引起的疾病和DDR2酪氨酸激酶的过度激活。 构成:修饰DDR2(盘状结构域受体)酪氨酸激酶活性的方法包括以下步骤:扩增编码含有DDR2激酶活性位点的氨基酸序列的cDNA,并将扩增的序列连接到谷胱甘肽-S- 转移酶(GST)产生DDR2激酶-GST融合基因; 将DDR2激酶-GST融合基因克隆到表达载体中,在宿主细胞中表达DDR2蛋白-GST融合蛋白的酪氨酸激酶活性位点,并纯化该蛋白; 并向融合蛋白中加入氧化应激以磷酸化DDR蛋白酪氨酸激酶活性位点中的酪氨酸。
