    • PURPOSE: A transgenic animal with genes relating to Alzheimer's disease and a production method thereof are provided, thereby the transgenic animal can be useful in studies on and development of medicines for Alzheimer disease. CONSTITUTION: An expression cassette for producing the transgenic animal with genes relating to Alzheimer's disease comprises (a) a mutated gene of amyloid precursor protein(APP), (b) a mutated gene of presenilin 1(PS1) and (c) a neuron specific promoter capable of regulating the said two mutated genes, wherein the promoter is neuron specific enolase(NSE); and an internal ribosomal entry site(IRES) may be inserted between the APP mutant gene and PS1 mutant gene. An expression vector containing the said expression cassette is constructed. The method for producing transgenic animal with genes relating to Alzheimer's disease comprises the steps of: constructing a vector containing NSE promoter and NSE intron 1; independently inserting a human APP mutant gene, a PS1 mutant gene and a BACE(β-site APP cleaving enzyme) gene into the vector to prepare three plasmids; isolating an insert to be inserted into an animal from the three plasmids; and injecting the insert into an abdominal cavity of an animal.
    • 目的:提供一种具有阿尔茨海默病相关基因的转基因动物及其生产方法,由此该转基因动物可用于阿尔茨海默病药物的研究和开发。 (a)淀粉状蛋白前体蛋白(APP)的突变基因,(b)早老蛋白1(PS1)的突变基因和(c)神经元特异性 能够调节所述两种突变基因的启动子,其中启动子是神经元特异性烯醇化酶(NSE); 并且可以在APP突变基因和PS1突变基因之间插入内部核糖体进入位点(IRES)。 构建含有所述表达盒的表达载体。 生产具有阿尔茨海默病相关基因的转基因动物的方法包括以下步骤:构建含有NSE启动子和NSE内含子1的载体; 将人APP突变基因,PS1突变基因和BACE(β-位点APP切割酶)基因独立插入载体中以制备三种质粒; 从三种质粒中分离插入动物的插入物; 并将插入物注射到动物的腹腔内。
