    • Dihydrodesoxystreptomycin (tertiary hydroxyl group of dihydrostreptomycin is reduced to hydrogen) is produced by reducing an aqueous solution of a streptomycin salt with amalgamated aluminium at a pH initially maintained at 1.5 to 2.4 and then at 2.7 to 3.5 when the reaction becomes vigorous as indicated by a rapid rise in temperature, which is preferably controlled by cooling to a temperature of 50 DEG C.-55 DEG C. On completion of the reaction (as indicated by the maltol test) the acid is neutralised by a base forming an insoluble salt with the acid, the precipitated salt so formed being removed and the free dihydrodesoxystreptomycin recovered by adding acetone or methyl alcohol. Specified streptomycin salts are the sulphate, hydrochloride-calcium chloride double salt and the pentachlorophenate-sulphate double salt. In the case of the pentachlorophenate-sulphate salt the reaction takes place in aqueous butanolic solution and the salt itself is prepared by adding streptomycin sulphate to sodium pentachlorophenate in methanol. Where the reaction takes place in the presence of (a) sulphuric acid, the sulphate ions are removed by the addition of barium hydroxide (b) hydrochloric acid, the chloride ions are removed by the addition of silver carbonate and the residual carbonate by the addition of barium hydroxide. Dihydrodesoxystreptomycin sulphate, nitrate, hydrochloride or phosphate is prepared by adding the appropriate acid to an aqueous solution of the base and precipitating the salt by the addition of methanol.
