    • The current invention reports a method for the cultivation of mammalian cells expressing a heterologous polypeptide, wherein a) the cultivation is performed in a cultivation vessel of a total volume of 25 ml to 3000 ml, b) the cultivation vessel contains a cultivation medium and an inside gas phase above the cultivation medium, c) the cultivation vessel contains a membrane or a cap or a lid separating said inside gas phase from the outside gas phase, d) in the cultivation vessel the agitation of the cultivation medium is performed by a mechanical movement of the entire cultivation vessel, e) during the cultivation the mechanical movement speed is changed depending on the viable cell density in the cultivation medium or it is changed depending on the oxygen partial pressure in the cultivation medium, f) the pH value of the cultivation medium is adjusted via the carbon dioxide concentration in the gas phase outside of the cultivation vessel, whereby the course of the viable cell density or the volumetric yield of the heterologous polypeptide expressed by the mammalian cell are similar to those obtained with a mechanically stirred cultivation vessel of a total volume of 1,000 1 to 25.000 1.
