    • A protein vaccine of avian influenza virus produced by using a reverse genetic engineering platform is provided to prevent virus diseases caused by various types of viruses by containing a conserved general immunogen, so that it is not necessary to develop a new vaccine when the virus type is changed by mutation. A method for preparing the protein vaccine of avian influenza virus comprises the steps of: (a) providing an amino acid sequence of at least one epitope peptide of a target antigen protein and converting the amino acid sequence into the corresponding wild type nucleotide sequence; (b) mutating the wild type nucleotide sequence of the epitope into the mutated nucleotide sequence which is recognizable by a host cell and encodes the epitope peptide; (c) synthesizing primers of the mutated nucleotide sequence; (d) synthesizing the mutated nucleic acid by using the primers in vitro; (e) linking the synthesized nucleic acid fragment to a plasmid and a nucleotide sequence having translocation and binding functions to produce the mutated plasmid; (f) transforming a host cell with the mutated plasmid to produce the epitope peptide encoded by the mutated nucleic acid; and (g) collecting and purifying the epitope peptide.
