    • PURPOSE: A method for producing foreign proteins by fed batch fermenting the mutant E. coli strain containing the modified oxygen dependent nar promoter is provided, therefore, the foreign proteins can be easily produced by regulating only the dissolved oxygen concentration. CONSTITUTION: The method for producing foreign proteins comprises the steps of: modifying the oxygen dependent inducible promoter nar to induce the expression of foreign proteins under microaerobic conditions; producing E. coli pMW618/narL containing the nar promoter; and incubating E. coli pMW618/narL under microaerobic conditions; lowering the dissolved oxygen concentration to 1 to 2%. Wherein, the microaerobic conditions is 20% or more of dissolved oxygen concentration and E. coli pMW618/narL is incubated in optical density of 20 to 80, at OD600.
    • 目的:提供通过补料分批发酵含有修饰的氧依赖性Nar启动子的突变型大肠杆菌菌株产生外源蛋白的方法,因此通过仅调节溶解氧浓度可以容易地产生外来蛋白质。 构成:产生外源蛋白质的方法包括以下步骤:修饰氧依赖性诱导型启动子,以在微需氧条件下诱导外源蛋白的表达; 生产含有纳尔启动子的大肠杆菌pMW618 / narL; 并在微需氧条件下培养大肠杆菌pMW618 / narL; 将溶解氧浓度降低至1〜2%。 其中,微生物条件为溶解氧浓度的20%以上,大肠杆菌pMW618 / narL的光密度为20〜80,OD 600。
