    • PURPOSE: Provided is a process for preparing a nucleocapsid(NC) protein of HIV in a mammalian cell easily. The nucleocapsid(NC) protein of HIV can be useful for development of anti-viral agent for treating HIV. CONSTITUTION: A process for preparing a nucleocapsid(NC) protein of HIV in a mammalian cell comprises the steps of: constructing an expression vector containing the HIV nucleocapsid protein gene, SV40 19s mRNA intron sequence or modified SV40 16s mRNA intron sequence positioned at the upstream of the NC gene, and CMV promoter; transforming the animal cell with the expression vector; culturing the transformed animal cell; and recovering the nucleocapsid(NC) protein of HIV, wherein the animal cell is COS-7 or 293T cell.

    • PURPOSE: Provided is a method for detecting an inhibitor of human immunodeficiency virus(HIV) using an E.coli strain JM109(KFCC-1159) cotransformed with vector pJC1 for the expression of HIV nucleocapside protein, and vector pNH1Psi(SL1234) including nucleotide sequence SL 1234(SEQ ID: NO.1) of HIV ψ and beta-galactosidase reporter gene(SEQ IN: NO.2). Therefore, it can be effectively used for the development of anti HIV inhibitor. CONSTITUTION: The detection method of HIV inhibitor comprises the steps of: culturing E.coli JM109(KFCC-11159) cotransformed with vector pJC1 and vector pNH1Psi(SL1234); treating the culture with a compound supposed to be an inhibitor of HIV; and measuring the expression level of beta-galactosidase in culture broth, decreased by the compound. The transformant characteristically decreases the expression of beta-galactosidase by linking nucleotide sequences of HIV protein and HIV ψ, and is thus used for the detection of HIV inhibitor.
