    • PROBLEM TO BE SOLVED: To construct a screening method for search of a substance specifically inhibiting the activity of a tRNA-dependent amidotransferase (GatCAB), to simply measure the deterioration of the GatCAB activity in which the construction of an enzymic reaction system such as substrate preparation is difficult, and to efficiently screen an enzyme-specific inhibitor expected as a medicament.
      SOLUTION: A coliform bacillus co-transformant of a plasmid starting transcription with a coliform bacillus RNA polymerase, and strictly controlling expression of a non-discriminating glutamyl-tRNA synthetase (ndGluRS) gene, and a plasmid permanently expressing GatCAB gene is constructed. The coliform bacillus is used as a test bacillus, and the deterioration of the GatCAB activity can readily be detected by measuring the growth of the coliform bacillus without preparing a substrate necessary for enzyme activity measurement and an enzyme protein. An inhibitor specific for the GatCAB can simply be searched for by screening a substance inhibiting the growth of the test bacillus without affecting a non-transformant.

    • PROBLEM TO BE SOLVED: To provide a simple inspection kit for xerostomia.
      SOLUTION: Determinate quantities of iodine and starch are spotted at specific positions of filter paper for paper chromatography, and a salivation stimulant, nicotine or capsaicin, is applied to an end of the filter paper to prepare filter paper for xerostomia diagnosis. The filter paper is put in the mouth of an examinee to cause the paper to absorb saliva. Thereafter, the amount of stimulus saliva secretion is measured from the position of a spot taking color with hydrogen peroxide solution. Or else, iron chloride (III) as a coloring agent is previously applied to the filter paper. The amount of stimulus secretion saliva is measured with capsaicin or nicotine applied to an end of test filter paper, the capsaicin or nicotine having a salivation stimulating action, allowing xerostomia to be diagnosed more surely. Further, determination is speeded and facilitated by using iron chloride (III) in place of hydrogen peroxide solution as a color-taking agent since coloration proceeds simultaneously with the absorption of saliva.
