    • PURPOSE: A recombinant Bacillus strain expressing cyclodextrinase and a method for preparation thereof by using recombinant vector pKYCD2 are provided. Therefore,cyclodextrinase having improved purity and homogenicity is produced massively at reduced cost. CONSTITUTION: The recombinant Bacillus strain, Bacillus polyfermenticus SCD pKYCD2(KCCM-10488) expresses cyclodextrinase. The preparation method of polyfermenticus SCD pKYCD2(KCCM-10488) comprises the steps of: constructing a recombinant vector pKY1 by cloning a vector p8A1 into a Bacillus-subtilis-E. coli shuttle vector pRB373; constructing a recombinant vector pKYCD1 by cloning a plasmid pBB702K containing a gene encoding cyclodextrinase derived from Bifidobacterium adolescentis lnt 57 into the recombinant vector pKY1; treating the recombinant vector pKYCD1 with restriction enzymes SmaI and EcoRV to prepare a recombinant vector pKYCD2; and transforming Bacillus polyfermenticus with the recombinant vector pKYCD2.

    • PURPOSE: Provided are a recombinant Bacillus strain which expresses β-xylosidase and a preparation method thereof by using a vector pKYXy12. Therefore, β-xylosidase having improved temperature stability and extended pH stability is mass-produced at reduced cost. CONSTITUTION: The recombinant Bacillus strain, Bacillus polyfermenticus SCD pKYXy12(KCCM-10487), expresses β-xylosidase. The method for preparation of Bacillus polyfermenticus SCD pKYXy12(KCCM-10487) comprises the steps of: constructing a recombinant vector pKY1 by cloning a vector p8A1 into a Bacillus-subtilis-E. coli shuttle vector pRB373; constructing a recombinant vector pKYXy11 by cloning a plasmid pXy11120 containing a gene which encodes cyclodextrinase derived from Bifidobacterium adolescentis lnt 57 into the recombinant vector pKY1; treating the recombinant vector pKYXy11 with restriction enzymes SmaI and EcoRV to prepare a recombinant vector pKYXy12; and transforming Bacillus polyfermenticus with the recombinant vector pKYXy12.
