    • PURPOSE: A recombinant Bacillus strain expressing cyclodextrinase and a method for preparation thereof by using recombinant vector pKYCD2 are provided. Therefore,cyclodextrinase having improved purity and homogenicity is produced massively at reduced cost. CONSTITUTION: The recombinant Bacillus strain, Bacillus polyfermenticus SCD pKYCD2(KCCM-10488) expresses cyclodextrinase. The preparation method of polyfermenticus SCD pKYCD2(KCCM-10488) comprises the steps of: constructing a recombinant vector pKY1 by cloning a vector p8A1 into a Bacillus-subtilis-E. coli shuttle vector pRB373; constructing a recombinant vector pKYCD1 by cloning a plasmid pBB702K containing a gene encoding cyclodextrinase derived from Bifidobacterium adolescentis lnt 57 into the recombinant vector pKY1; treating the recombinant vector pKYCD1 with restriction enzymes SmaI and EcoRV to prepare a recombinant vector pKYCD2; and transforming Bacillus polyfermenticus with the recombinant vector pKYCD2.

    • PURPOSE: Bacteriocin isolated from human skin and the use as a therapeutic agent for an acne are provided. The bacteriocin has wide antimicrobial activity, thermostability, pH stability and organic solvent stability, and is thus useful for the control of Propionibacterium acnes causing acne. CONSTITUTION: A microorganism Enterococcus faecium HJ35(KCCM 10465) produces bacteriocin of enterocin HJ35. An anti-acne composition comprises 0.1 to 20 wt.% of enterocin HJ35. A method for mass-producing enterocin HJ35 comprises the steps of: inoculating 1%(v/v) of Enterococcus faecium HJ35(KCCM 10465) into NLB medium; anaerobically fermenting the microorganism at 32 deg. C and 150 rpm; and adding 10 g/l(w/v) of glucose and 6 g/l(w/v) of lactic acid into the medium 2 days after fermentation, and culturing the medium.

CONSTITUTION: A method for improving the productivity and viability of Bacillus polyfermenticus SCD by alginate/methyl-cellulose encapsulation comprises the steps of: centrifuging a Bacillus polyfermenticus SCD cultured medium to obtain the pellet of Bacillus polyfermenticus SCD; washing and suspending the pellet to prepare the suspension; mixing the suspension of Bacillus polyfermenticus SCD with a mixed solution of sodium alginate and methyl-cellulose; and dropping the mixture into a 0.1M calcium chloride solution using a capsule-forming device to prepare Bacillus polyfermenticus SCD-encapsulating sodium-alginate capsules, wherein the sodium-alginate capsules have spherical shape with multipore and the diameter of 0.5 to 3.0mm; and the sodium-alginate capsules contain 2 to 4% of sodium-alginate and 0.5% of methyl-cellulose.
