摘要： A method and apparatus for measuring electrochemiluminescence from a sample composition are described wherein magnetically responsive electrochemiluminescent active species are captured on the electrode with the aid of a capture magnet having a configuration such that the magnetic flux lines (or the magnetic field gradient) of at least one magnetic field source therein are compressed and/or dispersed. This capture magnet improves the distribution of the magnetically responsive electrochemiluminescent active species on the electrode surface and reduces interference with the photomultiplier tube, thereby enhancing the ECL signal and improving sensitivity. The improved capture and distribution also allows for shorter assay times.

摘要： Bipyridine or phenanthroline ligands presenting functional groups that prevent non-specific binding (in particular, negatively charged functional groups that are unaffected by standard conditions for conjugating biological reagents through amide bonds) are described as are luminescent metal complexes comprising these ligands. The use of luminescent ruthenium and osmium complexes comprising these ligands in electrochemiluminescence assays shows that the use of these labels can significantly reduce the amount of non-specific binding observed relative to assays carried out using reagents labeled with analogous labels that don't present functional groups that decrease non-specific binding.

The apparatus may be used in an electrochemiluminescence assay for an analyte of interest in which magnetically responsive particles are magnetically collected on the electrode surface and emitted electrochemiluminescence is measured using the light detector.
