    • A transgenic, non-human mammal (in particular a mouse) is disclosed, whose body and sexual cells contain an additional DNA repair gene parasexually transmitted to the original animal at the 1-8 cell stage of embryonary development. The invention finds applications in the medical field, in particular the prevention and therapy of malign diseases, and in the pharmaceutical industry. The object of the invention is to provide a transgenic animal suitable for carrying out detailed research on the significance of cellular defence mechanisms and specific DNA-damages during tumour formation. This should give a new impulsion to the prevention and therapy of malign diseases. For this purpose, a transgenic, non-human mammal is provided which contains an additional DNA repair gene coding for a function suitable for eliminating alkylated DNA lesions, the coded function being an O -methylguanine-DNA-methyltransferase (MGMT), or for a function that corrects UV-induced damages. In a specially preferred embodiment of the invention, an additional recombinant repair gene is inserted which is expressed in the epidermis of the claimed mammal.

The proposed test procedure is characterised in that stable transgenic ES or EG cell clones containing tissue-specific promoters and reporter genes are selected, differentiation-dependent expression of tissue-specific genes is carried out following differentiation of ES cells in the presence of embryotoxic substances acting at specific times into different germination path derivatives; this is followed by detection of chemically induced activation, repression or modulation of tissue-specific genes which regulate embryonic development.
