摘要： A data storage apparatus includes an array of optical fibers. The array has a first end and a second end. The first end of the array includes multiple optical fiber ends, each optical fiber end having an end face adapted for receiving light of a wavelength λ into the fiber for conveyance to the second end of the fiber array. The second end of the array includes multiple tapered optical fiber tips, each tapered optical fiber end having a minimum diameter less than λ. An opaque coating covers a portion of the tapered optical fiber tips. The data storage apparatus also includes a photochromic medium located within a distance λ of the second end of the array.

摘要： The present invention provides biosensors, apparatus and methods for selectively detecting at least one complementary oligonucleotide target specie in a fluid sample containing a mixture of different oligonucleotide fragments. One preferred embodiment of the biosensor is as a unitary fiber optic array having an in-situ hybridization zone comprising not less than one specie of single stranded oligonucleotide disposed as individual deposits in aligned organization upon multiple strand end faces at differing spatial positions on the distal array end surface. In this manner, a collective of deployed, single specie, multiple fixed probes are presented for selective in-situ hybridization on-demand with at least one mobile complementary target specie ultimately bearing a joined identifying label. The biosensor provides for optical detection of in-situ hybridization on the distal end surface via the presence of the concomitantly disposed joined identifying label at the differing spatial positions.

摘要： The present invention relates to methods, systems, and kits for detecting, quantifying and/or analyzing a fluid sample comprising molecules or particles at low concentration. In certain embodiments, the methods for detection and/or quantifying analyte molecules in a sample comprise capturing a plurality of analyte molecules on a substrate (e.g., an array comprising a plurality of reaction vessels). The substrate may then be exposed to additional reaction components such as at least one binding ligand. The substrate may additionally be exposed to a precursor labeling agent molecule, wherein the precursor labeling agent molecule, in some cases, is converted to a labeling agent molecule, which may be detected, either directly or indirectly, which determination may be related to the presence of and/or may be employed to quantify the analyte molecules. Although the various aspects of the present invention may use a number of different assay formats, in one embodiment, the assays are conducted in a plurality of reaction vessels defined, at least in part, by the distal ends of fiber optic strands.

摘要： The present invention relates to a method and system of array imaging that extends or maximizes the longevity of the sensor array by minimizing the effects of photobleaching. The imaging system has a light source, a variable exposure aperture, and a variable filter system. The system extends the longevity of sensors by (1) using the variable exposure aperture to selectively expose sections of the sensor array containing representative numbers of each type of sensor, and/or (2) using the variable filter system to control the intensity of the excitation light, providing only the intensity required to induce the appropriate excitation and increasing that intensity over time as necessary to counteract the effects of photobleaching.

摘要： A microsphere-based analytic chemistry system and method for making the same is disclosed in which microspheres or particles carrying bioactive agents may be combined randomly or in ordered fashion and dispersed on a substrate to form an array while maintaining the ability to identify the location of bioactive agents and particles within the array using an optically interrogatable, optical signature encoding scheme. A wide variety of modified substrates may be employed which provide either discrete or non-discrete sites for accomodating the microspheres in either random or patterned distributions. The substrates may be constructed from a variety of materials to form either two-dimensional or three-dimensional configurations. In a preferred embodiment, a modified fiber optic bundle or array is employed as a substrate to produce a high density array. The disclosed system and method have utility for detecting target analytes and screening large libraries of bioactive agent.

摘要： A combinatorial synthesis method for fabricating unique families of discrete copolymer sensors and copolymer gradient sensors is provided. The method employs combinatorial copolymer synthesis of discrete monomer or oligomer combinations as well as spatially-varying combinations for generating large numbers of analyte-discriminating sensors from a limited selection of initial monomer and oligomer compositions. The method can be applied to either analyte-specific sensors or sensor arrays or semi-selective sensors and cross-reactive sensor arrays which employ virtually any known physicochemical transduction mechanism for detecting analytes. Since the analyte response characteristics of such copolymer sensors are not limited to a linear proportional ratio of the monomer or oligomer combinations employed, the resulting copolymer sensors provide for increased diversity in sensor and sensor array response characteristics for discriminating between a variety of materials and for detecting and identifying analytes in fluid samples.
