摘要:
PURPOSE: A high sensitivity real time confocal fluorescence microscope is provided to observe a single fluorescent molecular signal in real time. CONSTITUTION: A real time linear confocal microscope includes a light source unit(201), a lens unit(4), a first scan mirror(21), a microscope unit(2) and a detection unit(3). The light source unit consists of more than a short wavelength laser and collimates light by including more than one dichroic mirror. The lens unit includes more than one cylindrical lens and more than one circular lens, making the light of the light source unit into a linear form. The first scan mirror changes an angle, moves light to a direction perpendicular to the linear light and obtain the two dimensional fluorescence information of a target object. A multi-color mirror is placed in between the light source unit and the first scan mirror. The microscope unit projects light to the target object and condenses fluorescence generated from the target object. The detection unit includes a large area camera observing the fluorescence and a slit.
