摘要:
PURPOSE: A novel method for searching and quantitating target enzyme activation is provided to quickly sense and quantitate useful genes with high sensitivity. CONSTITUTION: A method for searching target enzyme activity using a genetic circuit comprises: a step of preparing a phenol compound sensing artificial genetic circuit containing a gene encoding phenol compound lyase control protein, reporter gene and gene expression control region; a step of preparing clone or gene library having a gene encoding detection enzyme; a step of introducing the artificial genetic circuit to a host microorganism; a step of treating the recombinant microorganism with phenol releasing-compound; and a step of sensing activation of induced reporter protein.

目的:提供一种新颖的方法,用于搜索和定量靶酶活化,以快速感测和定量高灵敏度的有用基因。 构成:使用遗传回路搜索靶酶活性的方法包括:制备含有编码酚化合物裂解酶控制蛋白基因,报告基因和基因表达控制区的基因的人工遗传回路的酚类化合物的步骤; 制备具有编码检测酶的基因的克隆或基因文库的步骤; 将人造遗传回路引入宿主微生物的步骤; 用酚释放化合物处理重组微生物的步骤; 以及感测报道蛋白激活的步骤。
