The invention relates to a diphosphonate derivative and application of composition containing the diphosphonate derivative to treat bone fracture. The structure formula of the diphosphonate derivative is shown in a formula I, wherein R1 is O(CH2)-A; A is an imidazole ring or a quinoline ring; R2 is hydroxyl or chloride. The diphosphonate derivative is used as a medicine for treating the bone fracture, has high affinity with hydroxyapatite in bone, and can form a chelating matter with calcium iron to be adsorbed onto the surface of the bone inorganic matter, so as to promote the withering of bone cells, and simulate the proliferation and maturing of the bone forming cells, thereby increasing the bone density. The diphosphonate derivative and the composition containing the diphosphonate derivative have the advantages that the function of promoting the bone healing is obvious, the treatment cycle is short, the side effect is small, the cost is low, and the diphosphonate derivative is suitable for being clinically popularized.
