    • A method for the modification of a specific gene of interest is described. The method employs two genetic constructs. One construct is a modification plasmid comprising the target gene or nucleic acid fragment of interest. The second construct is a modification cassette comprising yeast selectable markers, and optionally other DNA, bounded by flanking DNA which is complementary to portions of the target gene residing on the modification plasmid. Both constructs are transformed into budding yeast where homologous recombination occurs between the cassette and the plasmid. The resulting altered modification plasmid contains an insertion in the target gene in the regions governed by the sequence of the cassette flanking DNA.

    • Vectors which are to be integrated onto yeast chromosomes by multi-copying; and vectors which are to be integrated onto yeast chromosomes by multi-copying and show improved stability of expression units on the chromosomes. These vectors are ones each containing a marker gene for selecting a transformant, a shortened promoter sequence linked to the marker gene to thereby serve as an operator, and a sequence homologous with the chromosomal DNA of Candida utilis, optionally together with a heterogenous gene or a gene originating in C. utilis. It is linearized by cleaving with restriction enzymes within the homologous DNA sequence or at both ends of the same and, thus, enables the integration of the heterogenous gene or the gene originating in C. utilis onto the chromosomal DNA of C. utilis via homologous recombination.

    • The instant invention relates to methods for the in vivo assembly and integration of a polynucleotide of interest at a specific chromosomal target site in a fungal host cell, said method comprising transforming the host cell with: iv. at least one polynucleotide fragment comprising an upstream flanking region and a 5' part of the polynucleotide of interest in that order; v. at least one polynucleotide fragment comprising a 3' part of the polynucleotide of interest and a downstream flanking region in that order; and, optionally vi. one or more additional polynucleotide fragments each comprising a 5' part and a 3' part of the polynucleotide of interest; wherein the 5' part and 3' part of each polynucleotide fragment overlap with that or those of another fragment with at least 20 bp, whereby the fragments cover the entire polynucleotide of interest, and wherein the flanking regions are of sufficient size and homology to effectuate homologous recombination with the specific target site for chromosomal integration.
