    • PURPOSE: Transgenic cloned cow producing human erythropoietin and a method for producing the same transgenic cloned cow are provided, which method allows economic and effective production of various biomedicines, and facilitates production of human erythropoietin in cow's milk. CONSTITUTION: The nuclear transfer embryo produced by a fusion of a cow-derived enucleated oocyte with a nucleus of a cow-derived somatic cell transfected with a gene encoding human erythropoietin is provided, wherein the cow is an adult; the nuclear transfer embryo is SNU-B4(KCTC 10450BP). The method for producing a transgenic cloned cow expressing human erythropoietin in the mammary gland comprises the steps of: (a) introducing a gene encoding human erythropoietin into a cow-derived somatic cell line to prepare a nuclear donor cell; (b) removing cumulus cells surrounding a recipient oocyte and then the cytoplasm including the first polar body from the oocyte to prepare a cow-derived enucleated recipient oocyte; (c) transferring the transfected nuclear donor cell into the enucleated recipient oocyte and carrying out cell fusion to generate nuclear transfer embryos; and (d) transplanting the nuclear transfer embryo into a surrogate mother cow to produce live offspring.

    • PURPOSE: A method for inactivating nuclear chromosomal materials of recipient oocyte for manufacturing clone embryo from somatic cell using X-ray irradiation is provided, thereby increasing a blastocyst development rate and replacing the physical nucleus removing method. CONSTITUTION: The method for inactivating nuclear chromosomal materials of recipient oocyte for manufacturing clone embryo from somatic cell comprises irradiating X-ray to the recipient oocyte. The method for producing somatic cell cloned animals comprises the steps of: (a) irradiating X-ray to the recipient oocyte; (b) transplanting the somatic cell donor nucleus into the recipient oocyte; (c) inducing the re-programming of the nucleus transplanted somatic cell cloned embryo; (d) inducing the activity of the re-programmed somatic cell cloned embryo; (e) inducing the development of blastocyst of the activated somatic cell cloned embryo; and (f) inducing the implantation and generation of blastocyst, wherein the X-ray irradiation is carried out at 15-18 kVp; the X-ray irradiation is carried out at 2-8 mA; and the X-ray irradiation is carried out for 10-60 seconds.

    • PURPOSE: A transgenic cloned pig producing human erythropoietin and a method for producing the same transgenic cloned pig are provided, thereby producing a large quantity of human erythropoietin in the urinary bladder of the transgenic cloned pig with reduced costs. The transgenic cloned pig expresses human erythropoietin in the urinary bladder of pig. The method for producing a transgenic cloned pig producing human erythropoietin comprises the steps of: (a) transfecting and targeting a gene encoding human erythropoietin into a pig-derived somatic cell line to prepare a nuclear donor cell; (b) removing cumulus cells surrounding a recipient oocyte and then the cytoplasm including the first polar body from the oocyte to prepare a pig-derived enucleated recipient oocyte; (c) transferring the transfected nuclear donor cell into the enucleated recipient oocyte and carrying out cell fusion to generate nuclear transfer embryos; and (d) transplanting the nuclear transfer embryo into a surrogate mother pig to produce live offspring.

    • PURPOSE: A method for production of cloned animal embryos by nuclear transfer is provided to reduce the number of cloning processes. Therefore, itt can be useful for preservation of animals threatened with extinction, production of useful proteins, development of disease model, and supply of deficient organs. CONSTITUTION: The method for production of cloned animal embryos by nuclear transfer comprises the steps of: (i) inhaling a first polar body and 10 to 20% of cytoplasm without dying the nuclear of an egg cell by using a glass pipe having diameter of 20 to 40 micrometer to remove the nuclear from the egg cell; (ii) introducing a somatic cell into the first polar body and nuclear removed egg cell to prepare an egg-somatic cell complex within 1 minute during the inhalation process; and (iii) subjecting the egg-somatic cell complex to electric stimulation of 0.5 to 2 kv/cm and 6 to 150 microseconds 1 to 3 times to fuse and activate the egg-somatic cell complex, wherein the egg cell is collected from a mammal selected from pig, cow, sheet, mouse, rat, rabbit, dog and cat.
