    • 본발명은산꼬리풀추출물로부터분리된신규화합물, 상기추출물로부터분리된 (3R,5S,5aS,6R,7S,8R,8aS)-8-클로로-8a-히드록시-5-(2-히드록시아세톡시)헥사히드로-1H-3,6-메타노시클로펜타[e][1,3]디옥세핀-7-일 3,4-디히드록시벤조에이트{(3R,5S,5aS,6R,7S,8R,8aS)-8-chloro-8a-hydroxy-5-(2-hydroxy acetoxy)hexahydro-1H-3,6-methanocyclopenta[e][1,3]dioxepin-7-yl 3,4-dihydroxybenzoate} (KS-534) 을유효성분으로포함하는알러지, 염증, 천식또는만성폐쇄성폐질환의예방또는치료용조성물에관한것이다. 본발명에따른화합물들이 HT1080, H292 또는 EL4 세포에대한세포독성실험, TNF-α에의해유도된 MUC5AC의발현에미치는영향실험, NF-κB 루시페라제 (Luciferase) 리포터(reporter) 어세이 (assay), NF-κB 전사인자의활성저해효과실험(실험예 1)을통하여, NF-κB 활성억제를통한목표유전자 (MMP-9, MUC5AC 및 IL-4)의 mRNA 발현억제, NF-κb전사인자활성억제현상을통하여천식및 COPD를포함한폐질환치료에유용함을확인하였으며, MUC5AC 단백질생성에대한저해효과실험을통하여농도의존적으로 MUC5AC 생성을억제함을확인하였으며 (실험예 2); 난백알부민을이용한천식유발실험동물에서의항천식효과실험을통하여호산구를비롯한염증세포의수의현저한감소현상; 인터루킨-4, 인터루킨-5 와 13 수준의현저한감소효과, IgE의현저한감소, 염증세포의침윤의감소, 기관지상피의배상세포의점액분비의현저한감소현상등을통하여, 천식예방및 치료효과를확인하였으며 (실험예3), C57BL/6N 생쥐를이용한 COPD (만성폐쇄성폐질환) 모델에서 B기관지폐포세척액내 염증세포의수를현저하게감소함을확인하였으며, 활성산소족의생성억제, 호중구엘라스타제 (elastase) 활성억제, IL-6, TNF-a 수준의감소, 염증세포의침윤감소현상등을확인함으로서 (실험예 4) 만성폐쇄성폐질환의예방또는치료에유용하게사용될수 있음을확인함으로써, 상기조성물을알러지, 염증, 천식또는만성폐쇄성폐질환의예방및 치료용약학조성물, 건강기능식품또는건강보조식품으로유용하다.

    • A process for the isolation of a low molecular weight aminoglycan compound of formula I from a natural source of waste egg shells which is made up of alternating glucuronic acid and N-acetyl glucosamine units, wherein M may be at one or more instances Na, Ca, K, Mg; and n is an integer between 20 and 40 said process comprising the steps of: (a) pre-preparation of the waste egg shells for extraction of the embryonic low molecular weight aminoglycan compound of formula I using a polar organic solvent in water, (b) extraction of the low molecular weight aminoglycan compound of formula I as its water soluble salt using an aqueous polar salt solution, (c) isolation of a purified low molecular weight aminoglycan compound of formula I by gel formation out of the aqueous salt mixture by using a polar organic solvent followed by filtration or centrifuging, (d) stabilization of the isolated aminoglycan extract by sequential introduction of organic oils into a semi-dried gel to form aminoglycan compound of formula I. ® KIPO & WIPO 2009
