    • PURPOSE: A dual specificity oligonucleotide containing three different Tm sites is provided to overcome the problems and weaknesses of various methods including nucleic acid hybridization and to perform high specificity template-dependent reaction. CONSTITUTION: A dual specificity oligonucleotide to a target nucleic acid sequence is denoted by general formula(5'-Xp-Yq-Zr-3'). A universal base is deoxyinosine, 1-(2'-deoxy beta-D-ribofuranosyl)-3-nitropyrrole, or 5-nitroindole. A 3'-low Tm-specific site has a completely complementary and hybridizable nucleotide sequence to a template DNA. [Reference numerals] (a,DD) No expansion; (AA) Enhanced hybridization specificity of dual specificity oligonucleotide; (b,GG) No annealing; (BB) No expansion if a 5'-end part is annealed with a template instead of a 3'-end part; (c,JJ) Expanding; (CC,FF) With a template instead of a 3'-end part; (EE) No annealing if a 5'-end part has complementary sequence at a no-target position instead of a 3'-end part; (HH) Annealing 5'- and 3'- end parts with target sequences and successively expanding; (II) Target position

CONSTITUTION: A method for classifying and detecting sepsis-inducing microorganism comprises: a step of collecting nucleic acid molecule from a biological sample; a step of contacting primer pair for classifying microorganism group to nucleic acid molecule; a step of amplifying nucleic acid molecule obtained from biological sample through multiplex method; and a step of classifying and detecting sepsis-inducing microorganism. The biological sample is blood, whole blood, or serum.
