    • PURPOSE: An extension improving device of target molecules and a method thereof are provided to enhance stretching of the target molecule and to be used for detecting the target molecule at high throughput or analyzing the base sequence of nucleic acid. CONSTITUTION: An extension improving device(100) of target molecules comprises a first substrate(10), an ion selective transparent membrane prepared on the first substrate(30), and a second substrate(20) which includes one or more nano-channels(25). The first substrate is formed with transparent materials. The ion selective transparent membrane transmits cations and not transmits negative ions. The ion selective transparent membrane is formed with Nafion. A part of nano-channel is installed on the ion selective membrane, and the nano-channel and ion selective transparent membrane are crossed each other. The second substrate additionally includes an inlet port(21) in which a solution including target molecules is flowed and an outlet port(23) in which the solution is flowed out. The second substrate additionally includes one or more ion storages (31, 33) installed on the ion selective transparent membrane. In the nano-channel, the ion selective transparent membrane is formed in the inner wall.
