    • PROBLEM TO BE SOLVED: To provide a method for obtaining a highly specific and tailored ligand suitable for purifying a particular product target or eliminating a particular target impurity in a feed stream. SOLUTION: A designed affinity ligand binds the target with high specificity at a preselected binding condition and releases the target at a preselected elution condition. The ligand is isolated by bringing the target into contact with various polypeptides derived through diversification of the structure of a candidate binding domain, and variants (or analogues) including polypeptides favoring binding to the target under desired binding conditions and releasing from the target under elution conditions are included, wherein the binding and elution conditions differ depending on one or more parameters such as pH, temperature, salt concentration and volume% of an organic solvent.
