摘要： PROBLEM TO BE SOLVED: To provide an assay device.SOLUTION: The assay device includes: a liquid sample block; a reagent block located downstream having a reagent cell with a symmetrical line in the flow direction of a fluid in fluid communication with the sample block; reagent materials in a reagent cell including a first reagent material which is bilaterally symmetrical located at the symmetrical axis, and second and third reagent materials at mirror locations from the symmetrical line having substantially the same shape and volume; a detection block in fluid communication with the reagent block; and a suction block in fluid communication with the detection block having a capacity for receiving a liquid sample flowing out of the detection block. A sample addition block, the detection block and the suction block define a fluid channel.

摘要： PROBLEM TO BE SOLVED: To disclose a use of reaction kinetics.SOLUTION: This use of reaction kinetics is disclosed to generate multiple dose-response curves from a single reaction, thus eliminating the need to run a second experiment with additional sample, reagents, and time to cover a broader measuring range than is available in a standard assay. Using a single protocol, the differences in the reaction kinetics for different sample concentrations yield different responses at different measurement times. Selection of an appropriate dose-response curve cross-section increases the measuring range and accuracy of the assay from a single reaction without substantially increasing imprecision. Several overlapping dose-response curves are pieced together to provide a standard curve to ensure continuity throughout the expanded measuring range.

The detection zone 40 has a substrate and projections which extend substantially vertically from the substrate, where the projections have a height, a cross section and a distance between one another that defines a capillary space between the projections capable of generating a capillary flow parallel to the substrate surface. The device is capable of creating a reagent plume in the detection zone 40 that includes a liquid sample and a dissolved reagent, where the width of the reagent plume extends substantially across the width of the detection zone 40. The device further includes a wicking zone in fluid communication with the detection zone 40 having a capacity to receive the liquid sample flowing from the detection zone 40.

The reagent cells divide a sample flow from the sample zone into multiple flow streams. Also included are: one or more flow control elements disposed downstream of the reagent zone and combining the multiple flow streams into fewer flow streams; a detection zone 40 in fluid communication with the reagent zone; and a wicking zone 50 in fluid communication with the detection zone having a capacity to receive a liquid sample flowing from the detection zone. The sample addition zone, the detection zone and the wicking zone define a fluid flow path.
