    • PROBLEM TO BE SOLVED: To provide a TuSV (tulip streak virus) protein, a nucleic acid encoding the protein and a nucleic acid which is a complementary chain to the nucleic acid. SOLUTION: A coat protein of a refined TuSV is purified therefrom and the partial amino acid sequence thereof is determined. A DNA coding the coat protein of the TuSV is amplified by an RT-PCR (reverse transcription polymerase chain reaction) using a primer designed on the basis of information about the determined amino acid sequence. The amplified DNA is cloned to determine the primary structure thereof. A RACE (rapid amplification of cDNA ends) method using a primer designed on the basis of the resultant sequence information is carried out to thereby succeed in isolating the DNA encoding the coat protein of the TuSV and determining the primary structure thereof. A TuSV-resistant plant can be created and the infection of the TuSV can be diagnosed by utilizing the DNA. COPYRIGHT: (C)2005,JPO&NCIPI
