摘要： Multi-step systems and methods for target-molecule purification that employ column chromatography-based or membrane filtration- based polishing steps are disclosed. A target protein-containing eluate having a high residual salt concentration is collected from a first chromatography column prepared with an affinity chromatography resin, loaded onto a second chromatography column prepared with cation-exchange resin, and eluted from the cation-exchange column using a buffer in which a time-dependent pH gradient is established. A partially purified target protein-containing eluate may also be further purified by passing the target protein-containing eluate through a salt-tolerant anion exchanger.

摘要： A method of conducting an enzymatic process in which the enzymes are recovered and reused in at least a second iteration of the process, wherein each iteration of the process comprises the steps of: (a) providing a heterogeneous substrate solution, in which the substrate is fully soluble, partially soluble or insoluble; (b) adding an enzyme or mixture of enzymes to the heterogeneous substrate solution; and (c) allowing an enzymatic reaction of the substrate to proceed; wherein, after completion of step (c) for each iteration, the enzyme is recovered from the mixture resulting from step (c) according to the following steps: (d) conducting a non-packed-bed adsorption process comprising contacting the reaction mixture with an adsorbent that adsorbs the enzyme in order to separate the enzyme from the reaction mixture resulting from step (c); (e) optionally washing unbound material from the adsorbent; and (f) desorbing the enzyme from the adsorbent; and further wherein the desorbed enzyme obtained in step (f) is used in step (b) of at least one subsequent iteration of the process.

摘要： The present invention relates to a method for pre-fractionation of complex protein and/or peptide samples. The method comprises the following steps: a) loading the sample onto pre-packed media able to separate proteins/peptides according to their pI-value; b) eluting the media by centrifugal force under denaturing conditions with at least three buffers having different pH in a stepwise manner to obtain at least three fractions separated according to pI-value; and c) subjecting each fraction to further separation in a pH-range corresponding to the pI-values of said fractions. The invention also relates to a kit comprising at least one spin column or multiwell plate filled with chromatography medium able to separate proteins/peptides according to pI-value, at least three buffers, and one or more IPG strips having a narrow pH-range adapted to each of said buffers.

The first eluent solution has a first pH and the second eluent solution has a second pH where the first pH and second pH are different values.
