摘要： PROBLEM TO BE SOLVED: To provide a vector construct useful in the expression of double-stranded RNA.SOLUTION: The DNA construct comprises (a) a first promoter and (b) a second promoter, wherein the first promoter and the second promoter are in opposite orientation to each other. Additionally the construct comprises (c) an inter-promoter region that is located downstream of the 3' terminal of the first promoter and downstream of the 3' terminal of the second promoter and defined, (d) at least one cloning site located in the inter-promoter region, and (e) a first transcription terminator which is located downstream of the first promoter and downstream of at least one cloning site when viewed from the 3' terminal side of the first promoter. The first transcription terminator is operably linked to the first promoter. The construct is useful for expression of double-stranded RNA in vitro and in vivo.

摘要： PROBLEM TO BE SOLVED: To provide an RNA interference method for controlling pest infestation using a double stranded RNA molecule that represses, delays or reduces the gene expression within a particular pest.SOLUTION: The double stranded ribonucleotide sequence produced from the expression of a polynucleotide sequence can inhibit the growth of a pest when in contact with the pest. Thus there are provided the double stranded ribonucleotide sequence, a composition including the polynucleotide, and cells transformed by the polynucleotide. Provided is a method for controlling the infestation of pest wherein the composition and the cells are selected from the group consisting of a spray, powder, pellet, gel, capsule, food product, garment bag, and book, and the food is selected from the group consisting of stored grains, pet food and powdered chocolate. Also provided is a method for preventing or treating the infestation of pest such as nematode and true fungi by treating the surface of an object such as wood, tree, book binding, cloth, food product and storage container with the composition.
