    • A signal processing method in autoradiography for determination of the base sequence of DNA or a DNA fragment, employing groups of radioactively labeled base specific cleavage products or mixtures thereof obtained by specifically cleaving the DNA or DNA fragment labeled with a radioactive element and resolved one-dimensionally in parallel relation to each other to form resolved rows on a support medium. An autograph is obtained having locational information on groups of radioactively labeled cleavage products contained in the rows on the support medium. An electrical digital signal corresponding to the autoradiograph is then generated by recording and reproducing from a stimulable phosphor sheet the radiation energy stored therein. Sampling points in each resolved row of the digital signal are next detected. Reference sampling points are then determined in a plurality of reference rows which are either directly provided on the support medium and/or synthesized from certain resolved rows. Sampling points in the remaining non-reference rows are identified by comparison with the continuous lines to thus determine the base sequence of the DNA or DNA fragment.

    • A signal detecting method in autoradiography, which is applied to an autoradiography having locational information on radioactively labeled substances such as radioactively labeled biopolymers distributed one-dimensionally, being recorded on a stimulable phosphor sheet as a radiation energy-stored image, which comprises steps of:(1) preliminarily scanning a part of the stimulable phosphor sheet with an electromagnetic wave to release a portion of the radiation energy as stimulated emission and detecting the stimulated emission to give an electric signal, to which a signal processing is applied, to determine one-dimensional distribution direction of the radioactively labeled substances; and(2) finally scanning the stimulable phosphor sheet with an electromagnetic wave along said one-dimensional distribution direction to release the radiation energy as stimulated emission and detecting the stimulated emission, to obtain the locational information on the radioactively labeled substances as a digital signal.

    • A signal processing method in autoradiography for obtaining information on the one dimensional location of radioactively labeled substances in a sample where the substances of the sample are distributed in at least one row on a support medium where the information is in the form of symbols, numerals or a combination thereof. The method at least includes the steps of (1) generating digital image data where the data comprises sets of digital signals corresponding to the autoradiograph including the row of distributed substances and where the sets of digital signals may respectively represent successive linear portions of the autoradiograph; (2) scanning selected digital signals of the digital image data to generate at least one further signal representative of a predetermined portion of the row of distributed substances where the position of the predetermined portion is at least partially determined by its location in the scanning direction and the amount of radioactively labeled substance at the location is at least partially determined by the amplitude of the further signal; (3) processing the further signal with either or both of smoothing and threshold-processing to detect sampling point corresponding to the location of the predetermined portion of the row of distributed substances; (4) obtaining the image data digital corresponding to the autoradiograph having the locational information on the radioactively labeled substances where the digital image data is obtained by utilizing a radiation image recording and reproducing method of employing a stimulable phosphor sheet. A signal processing method for determining the base sequence of DNA is also disclosed.

Sampling points in each resolved row of the digital signal are next detected. Reference sampling points are then determined in a plurality of reference rows which are either directly provided on the support medium and/or synthesized from certain resolved rows. Corresponding reference sampling points in the plural reference rows are joined to generate plural continuous lines comprising straight lines, polygonal lines or curved lines. Sampling points in the remaining non-reference rows are identified by comparison with the continuous lines to thus determine the base sequence of the DNA or DNA fragment.
