    • There is provided plantlet handing apparatus including a housing (70) for a nutrient medium. A plurality of plantlet holders (72) each include a pair of body portions forming, in assembly, an open-topped container having apertures open to the nutrient medium in use. The body portions include engagement means engaging the body portions in assembly for handling and permitting separation. A plantlet holder support (71) is located in the housing and supports the plantlet holders in the nutrient medium while maintaining the body portions together. A closure (73) has a substantially airtight seal to the housing and has windows (111) of selected permeability to metabolic gases produced or required by plantlets.

    • A method of modifying a plant to increase vegetative growth of commercially valuable plant structures at the expense of non-essential and non-commercial structures is provided. From reproductive tissues of both Pinus radiata and Eucalyptus grandis, cDNA were isolated corresponding to genes which were specifically expressed during early development in both male and female plant structures. The promoter regions to these genes were isolated from respective genomic DNAs and fused to a structural gene for a bacterial ribonuclease, Barnase. In a variation of the method, an expression control system was used to control promoter leakage. Gene constructs were also prepared which expressed anti-sense forms of essential MADS box genes for these plants. Candidate gene constructs were used to transform shoot-forming tissues of target eucalypt and pine species. Explants from successful transformants were assessed for incorporation of the gene constructs and production of sterile plants.

    • There is provided a method of regulating a eukaryotically active gene, comprising transforming a cell with a transformation cassette expressing a modulator gene product regulating the eukaryotically gene or its product and a further gene product regulating said modulator gene or its product, the promoters of two of said gene, modulator gene and further genes being selected from inducible promoters and developmental promoters for the same or complementary tissues. The lethal gene expressing barnase, a ribonuclease of B. amiloliquefaciens, is placed under the control of a tissue specific promoter, such as those derived from PrMADS1, 2 or 3 of Pinus radiata or EGM1, 2 or 3 of Eucalyptus grandis. The same tissue specific promoter is used to express Laclq gene, a repressor for barnase (barstar) being promoted by a modified 35s RNA CaMV promoter including the lac operon. The cassette is used to transform plant cells for regeneration into plants expressing the barnase in the target tissues with improved specificity and reduced promoter leakage.

    • A process for the optical resolution of racemic 3-(p-chlorophenyl)-glutaramide (GAM) into its R isomer R-COOH and its S isomer S-COOH, which process includes the steps of either: (1) reacting racemic 3-(p-chlorophenyl)-glutaramide dissolved in a suitable solvent with S-(-)- alpha -methylbenzylamine of the formula H2N-S'; (2) precipitating out of the solution of step (1) R-CO2 .H3 N-S'; (3) dissolving the precipitate of step (2) in water, with the addition of a suitable acid; and (4) precipitating out of the solution of step (3) R-COOH; or (5) reacting racemic -3-(p-chlorophenyl)-glutaramide dissolved in a suitable solvent with R-(+)- alpha -methylbenzylamine of the formula H2N-R'; (6) precipitating out of the solution of step (5) S-CO2 .H3 N-R'; (7) dissolving the precipitate of step (6) in water, with the addition of a suitable acid; and (8) precipitating out of the solution of step (7) S-COOH.
