摘要： The current disclosure provides novel methods and compositions to suppress immunity and are useful for treating autoimmune and inflammatory conditions, and in some circumstances, cancers. Accordingly, aspects of the disclosure relate to a method for treating an inflammatory, autoimmune, autoinflammatory or cancer disease or condition in a subject comprising administering a DUX4 polypeptide or nucleic acid encoding a DUX4 polypeptide to the subject. Further aspects relate to a method for treating a tissue transplant subject, the method comprising administering a DUX4 polypeptide or nucleic acid encoding a DUX4 polypeptide to the subject. The present inventions also relate to universal donor stem cells that overcome immune rejection in cell-based transplantation therapies.

摘要： The present disclosure provides compositions and methods for producing and expressing variant or engineered I-Onul endonucleases, variant or engineered I-Onul homologues, and hybrids of two I-Onul or I-Onul homologue domains that have a target site altered from the wild-type. 摘要： The present invention provides vaccine compositions for protection against human rotaviral disease designed for use in particular areas of the world. Human x bovine reassortant rotavirus comprising each of the four clinically most important VP7 serotypes of human rotavirus are combined with other VP7 serotypes typically found in the area of interest into a multivalent formulation which provides a high degree of infectivity and immunogenicity. Methods and an administration protocol for producing an immunogenic response without producing an increased risk of intussusception are also provided.

摘要： Virus-specific CD4 + T cell help and CD8 + cytotoxic T cell responses are critical for the maintenance of effective immunity in chronic viral infections. The importance of the CD4 + T cells has been documented in HIV infection. A T1-specific CD4 + T cell line from a healthy volunteer immunized with a canarypox vector expressing gpl20 has been developed. The cell line was restricted to DR13, which is common in the U.S. in both Caucasians and African-Americans and is one of the major haplotypes in Africans. Amino acid substitutions in the T1 epitope were made to induce a stronger epitope-specific CD4 + T cell response than the original epitope resulting in an improved CD4 epitope. A polypeptide comprising the enhanced CD4 epitope can be used as a component in compositions either alone or in combination with other adjuvants and other immunogenic compositions to induce a more effective immune response to HIV infection.

摘要： Disclosed are methods for sreening an agent for activity in modulating a T lymphocyte. The methods generally comprises contacting a HSV U s 3-expressin cell with a T lymphocyte in the presence or absence of the agent and monitoring a physiological change associated with CTL function to determine whether the agent modulates CTL activity. In particular, the HSV U s 3-expressing cell line can be a HSV-infected fibrolast and the physiological change associated with CTL function is a recduction or inhibition of the phosphorylation of HSP90 and/or a 50 kD CTL protein. Also disclosed are methods for blocking suppression of CTL activity against HSV-infected target cells, as well as methods for suppressing CTL activity against a target antigen.
