摘要： The human homologs of the yeast ubp8 gene and polypeptide are required for chromatin development and gene regulation of the genes in the SAGA complex, including H2B. Compounds that inhibit the expression or activity of the human Ubp8 homolog polypeptide and the nucleic acid sequences encoding it are useful in rendering a cancer cell more sensitive to additional anti-tumor therapies, in diagnosing tumorigenic cells, or in treating or diagnosing other disorders dependent upon regulation of genes requiring deubiquitylation for transcription.

摘要： An organotypic culture comprises an artificial stroma overlayed with epithelial cells isolated from a human colon or intestine. The stroma comprises a mixture of collagen and human fibroblasts isolated from a human colon or intestine. The culture contains a factor that binds the IGF-1 receptor, a factor that binds the EGF receptor, and a factor that binds the LIF receptor. These factors may be added exogenously to the culture via medium or may be expressed by various recombinantly engineered cell types in the culture. The organotypic culture can result in growth that is in situ -like or emphasizes other physiological or morphological states, depending on the balance of factors in the growth media. The organotypic culture may be used in methods for screening of therapeutic, carcinogenic, or growth enhancement factors, or for treating intestinal injuries by applying to the site of an injury the intact culture or the components thereof.

Also provided are methods of inducing interferon-α and interferon-β by delivering a recombinant simian adenovirus to a subject. The methods and compositions of the invention are particularly well suited for prophylaxis and treatment of infections with human immunodeficiency virus and human papilloma virus, among others, and cancer therapy.

摘要： Compounds useful in the treatment of infection by an episomal DNA virus are identified by a method involving the steps of (1) providing a culture of mammalian or avian cells containing multiple copies of a recombinant viral extrachromosomal episome having an episomal maintenance element, the episomes being associated with a detectable label; (2) contacting the culture with a test molecule that inhibits the interaction between the episomal maintenance element on the episomes and at least one enzyme selected from a family of enzymes consisting of the poly-ADP ribose polymerase family, the poly-ADP glycosylase family, the telomerase family, the myb DNA binding family that interferes with telomeres, and the telomere repeat binding protein family; and (3) detecting the elimination of the episomes from the cells by detecting the presence of the detectable label in the medium of the culture. These compositions are useful in pharmaceutical compositions for treating an infection by an episomal DNA virus and in diagnostic kits.

摘要： The present invention relates to a method for producing peptides which have biological activity comprising: (i) identifying a region of a protein of interest which is homologous to a member of the immunoglobulin superfamily of molecules; (ii) synthesizing a peptide which comprises amino acid sequence found in the region of homology; and (iii) modifying the synthetic peptide such that it assumes a three-dimensional conformation which is biologically active. The invention is based, in part, on the discovery that a cyclic peptide comprising amino acid sequences of a CDR derived from an antibody directed toward a viral receptor molecule can be used to effectively inhibit the binding of virus to its receptor. The invention is also based in part on the discovery that a cylic peptide corresponding to a portion of human immunodeficiency virus (HIV) gp120 that is homologous to immunoglobulins may be used to generate an anti-HIV immune response.
