    • PURPOSE:To optimize the setting of a base line by recording the output of a measuring instrument as it is and selecting a base line level from measurement record after measurement. CONSTITUTION:When the measurement is started, the measurement output is stored in a memory as it is. In this case, the measurement output is stored in the memory while the output of the measuring instrument which is 0 is regarded as level 0. After the measurement, the measurement data stored in the memory are processed. Namely, the measurement data in the memory 2 are read out and the coordinates of the start and end points of each peak are detected. Then the base line level is set. This base line level is set by a person who takes an analysis by displaying the measurement result. After the setting of this base line level, the base line is subtracted from the measurement data in the memory 2 and the resulting data are displayed on a display device 3; and it is confirmed visually whether or not the base line setting is proper and the peak area is calculated. Consequently, the base line is correctly set and the area of each peak can be found without remeasurement.

CONSTITUTION:The chromatogram waveform P to be subjected to data processing is assumed to be displayed. A peak number n=5 is inputted by a key at this time. The beginning point, vertex and end point of the first peak P0 are then assigned by a cursor and the vertexes and end points of the respective peaks up to the final peak P4 are successively assigned. The start point A of the first peak P0 and the end point B of the final peak P4 are connected and a base line q is determined when the above-mentioned assignment ends. A perpendicular (r) is drawn to the base line q from the end points of the respective peak values P0 to P4 and the peak division is executed. The area enclosed by this base line (q) and the perpendicular (r) is calculated.

On the other hand, the optical system 5 to change the incident direction of the exciting light 6 is inserted into the optical path of the exciting light 6 at the time of making fluorescent measurement. The incident direction of the exciting light 6 is made into the direction diagonal with the microplate and the exciting light 6 is no longer made incident to the detector 10 by inserting this optical system 5. The fluorescence 24 is released in the direction of the detector 10 as well as against this and, therefore, the fluorescence 24 is made incident to the detector 10 and is received therein. The transmitted fluorescence is measured in the state of the high S/N in this way.

When switches (SW) 24, 34 are opened in a section 8a, the photometric signal voltage and monitor signal voltage at the A-point are respectively held to a sample holding circuits 26, 36 and respective held voltages are supplied to a dynode feedback circuit until reaching the B-point. Succeedingly, when SW 44, 50 are connected to a b-contact side at the B-point, the difference between held voltage Vh and photometric voltage Vs is operated by the differential amplifying circuit 42 of a weighted averaging circuit 40 and multiplied by a constant (t) by an integrating circuit 46. Further, SW 24, 34 are closed between points B-C and SW 44, 50 are connected to an a-contact side to release sample holding. By this method, the dynode feedback circuit also operates smoothly.
