    • PURPOSE: The method for fixing oligonucleotides on the support and oligonucleotide array manufactured thereby are provided, therefore the pretreatment process for modification of oligonucleotides and supports is not required and the various kinds of oligonucleotides can be fixed simultaneously with high hybridization efficiency. CONSTITUTION: The method for fixing oligonucleotides on the support comprises the steps of: coating a hydrophobic fixing layer being solidified under the appropriate conditions on the support; dropping the droplets of oligonucleotide solution having a hydrophobic group at 3- or 5-end on the specific region of the fixing layer; and solidifying the fixing layer. Wherein, the oligonucleotides with the hydrophobic group are the PCR amplified products by using a primer having the hydrophobic group at its 5-end; the fixing layer is a polymer formed by polymerization or polycondensation; and the hydrophobic group is dimethoxytrithyl group. The alternative method for fixing oligonucleotides on the support comprises the steps of: fluidizing the solidified hydrophobic fixing layer, which is coated on the support, under the appropriate conditions; dropping the droplets of oligonucleotide solution having a hydrophobic group at 3- or 5-end of oligonucleotides on the specific region of the fixing layer; and solidifying the fixing layer.
    • 目的:提供在由其制备的载体和寡核苷酸阵列上固定寡核苷酸的方法,因此不需要修饰寡核苷酸和载体的预处理方法,可以以高杂交效率同时固定各种寡核苷酸。 构成:在载体上固定寡核苷酸的方法包括以下步骤:在适当的条件下将载体固化的疏水性固定层涂覆在载体上; 在固定层的特定区域上滴加在3或5端具有疏水基团的寡核苷酸溶液液滴; 并固化固定层。 其中具有疏水基团的寡核苷酸是通过在其5末端使用具有疏水基团的引物的PCR扩增产物; 固定层是通过聚合或缩聚形成的聚合物; 疏水基团为二甲氧基三甲苯基。 在载体上固定寡核苷酸的替代方法包括以下步骤:在适当的条件下将涂覆在载体上的凝固的疏水性固定层流化; 在固定层的特定区域上滴加在寡核苷酸的3或5端具有疏水基团的寡核苷酸溶液滴; 并固化固定层。
