摘要： PROBLEM TO BE SOLVED: To optimize the hydrocarbon decomposition rate of autochthonous microorganisms (petroleum decomposition bacteria) and to restore soil polluted with petroleum in an early period by mixing the soil polluted with the petroleum and nitrogen sources and phosphorus sources, then adjusting the air supply rate into the soil mixture and controlling the temp. in the soil mixture. SOLUTION: At the time of the restoration treatment of the soil polluted with the petroleum, the soil 3 polluted with the petroleum is first charged into a hopper 1 and is sent to a mixer 2 by a belt conveyor 4. An aq. soln. 5 contg., for example the nitrogen sources and phosphorus sources is supplied to this mixer 2 and is homogeneously and continuously mixed with the soil 3 polluted with the petroleum. After the completion of mixing, the restored soil 6 discharged from the mixer 2 is carried out by the belt conveyor 4. At the time of such treatment, the fresh air is supplied to the soil mixed with inorg. nutrients to decompose and remove the petroleums in the soil by the effect of the autochthonous microorganisms (petroleum decomposition bacteria). The decomposition rate of the petroleums is so controlled as to attain the range where the rate is highest by utilizing the decomposition reaction heat of the petroleum generated at this time.

摘要： PROBLEM TO BE SOLVED: To detect yeasts and actinomyces in natural and human environments in high sensitivity by using the DNA extracted from microorganisms in the environment through two specific washing treatment as a template, selecting a specific sequence as a primer and subjecting them to polymerase chain reaction. SOLUTION: An environment sample is subjected to the following two washing treatments: (A) the first washing treatment in which the sample is washed with a weak-acidic aqueous solution containing an inorganic or organic slat or an aqueous urea solution and (B) the second washing treatment in which the sample is washed with an aqueous solution of powered milk to extract DNA from the environmental microorganisms in the environmental sample. The extracted DNA is used as a template and the primers of formula I and formula II are used to effect PCR. Then, the amplified DNA fragment is hybridized with the DNA of formula III whereby the yeast in the environment can be detected. In the meantime, PCR is performed by using the primers of formula IV and formula V, the DNA of formula IV is used to effect hybridization whereby the actinomyces in the environment are detected.
