摘要： A process for modifying the surface of a silicon-containing polymer is presented, the process comprising the steps of: imbibing a zwitterionic monomer or a free radical initiator into a silicon-containing polymer in an imbibing mixture comprising the silicon-containing polymer, at least one of the zwitterionic monomer and the free radical initiator, and a solvent system; and polymerizing the zwitterionic monomer to modify the surface of the silicon-containing polymer with a zwitterionic polymer in a polymerization reaction mixture which comprises the imbibed silicon-containing polymer, a solvent system and the zwitterionic monomer or the free radical initiator, the polymerization reaction mixture having a composition that differs from the composition of the imbibing mixture. Such a process can be applied to prepare medical devices such as contact lens having a wettable, biocompatible surface. Processes for producing such devices are also described.

摘要： Substrates, optionally coated with an undercoating layer, having grafted there from one or more non-fouling materials are described herein. The non-fouling, polymeric material can be grafted from a variety of substrate materials, particularly polymeric substrates and/or polymeric undercoating layers. The graft-from techniques described herein can result in higher surface densities of the non-fouling material relative to graft-to formulations. Graft-from methods can be used to produce covalently tethered polymers. The compositions described herein are highly resistant protein absorption, particularly in complex media and retain a high degree of non-fouling activity over long periods of time. The compositions described herein may also demonstrate antimicrobial and/or anti-thrombogenic activity. The non-fouling material can be grafted from the substrate, or optionally from an undercoating layer on the substrate, preferably without significantly affecting the mechanical and/or physical properties of the substrate material.

摘要： Compositions containing one or more types of membrane-targeting antimicrobial agents immobilized on a substrate with activity in relevant biological environments, and methods of making and using thereof, are described herein. The antimicrobial agents retain their activity in the presence of blood proteins and/or in vivo due to improved molecular structures which allow for cooperative action of immobilized agents and hydrophilic chemistries which resist non-specific protein adsorption. Suitable molecular structures include branched structures, such as dendrimers and randomly branched polymers. The molecule structures may also include hydrophilic tethers which provide both flexibility and resistance to non-specific protein adsorption. The membrane targeting antimicrobial agent coatings can be applied to a variety of different types of substrates including medical implants such as vascular grafts, orthopedic devices, dialysis access grafts, and catheters; surgical tools, surgical garments; and bandages. The substrates can be composed of metallic materials, ceramics, polymers, fibers, inert materials such as silicon, and combinations thereof. The compositions described herein are substantially non-leaching, resistant to non-specific protein adsorption, and non-hemolytic.

摘要： Synthetic amino acids containing one or more non-fouling groups or moieties are described herein. In one embodiment, the amino acid has the following chemical formula: (I), where L is a linker group and Z is a non-fouling group including, but not limited to, polyethylene glycol (PEG); oligoethylene glycol (OEG); zwitterionic group, such as phosphorycholine, carboxybetaine, and sulfobetaine; groups that are hydrogen bond acceptors but not hydrogen bond donors. The non-fouling amino acids can be incorporated into a bioactive peptide as single amino acid residues, multiples amino acid residues, or as blocks of amino acids. The non-fouling amino acids, or peptides containing one or more non-fouling amino acids, can be applied to surfaces in order to improve biocompatibility, reduce thrombogenesis, and/or reduce fouling by proteins or bacteria present in solution.
