    • PURPOSE: Provided is a method for effectively transforming a Taxus cuspidata cell line by using Agrobacterium, thereby increasing transformation efficiency of plants more than two times and expressing a transformed gene stably to improve the production the second metabolites, such as paclitaxel. CONSTITUTION: The method comprises the steps of: pre-treating plant cells chemically and physically; co-culturing Agrobacterium transformed with a recombinant plasmid and plant cells in a medium of pH 5.0-5.6 containing pathogens, for 1-4 days; washing the culture then followed by removing Agrobacterium and culturing the plant cells in a selection medium; and selecting a tissue growing well to obtain a cell line which expresses a foreign gene stably.
    • 目的:提供一种利用土壤杆菌有效转化东北红豆杉细胞系,从而提高植物转化效率两倍以上并稳定表达转化基因以提高紫杉醇等第二代谢产物生产的方法。 组成:该方法包括以下步骤:化学和物理预处理植物细胞; 在含有病原体的pH 5.0-5.6的培养基中共培养用重组质粒和植物细胞转化的土壤杆菌1-4天; 洗涤培养物,然后除去农杆菌并在选择培养基中培养植物细胞; 并选择生长良好的组织以获得稳定表达外源基因的细胞系。

    • PURPOSE: A method for preparing a recombinant protein inhibiting angiogenesis using a geminivirus expression system is provided, thereby reducing the preparation costs and increasing yield in comparison with using a microorganism or animal cell expression system such as Escherichia coli or insect cell expression system. CONSTITUTION: The method for preparing a recombinant protein inhibiting angiogenesis using a geminivirus expression system comprises the steps of: constructing a recombinant vector containing two geminivirus replicons between two T-DNA boarders, and endostatin or tumstatin gene between the geminivirus replicons; transforming a microorganism with the recombinant vector; transfecting a plant tissue or plant with the transformed microorganism; and culturing the transfected plant tissue or plant in a medium.
