    • The present invention provides methods and systems that uses transgenic zebrafish with an easily assessable reporter gene under the control of pollutant-inducible DNA response elements. Transgenic zebrafish, carrying pollution-inducible response elements, are placed in the water to be tested, and the contaminants become bioconcentrated (generally 1,000- to 40,000-fold, relative to the water) in the tissues of the fish thereby activating specific response elements, which up-regulate the LUC reporter gene. Fish are then removed from the test water and placed immediately in a luminometer cuvette and incubated with luciferin. Luciferin is rapidly taken up into the tissues of the fish, oxidized by luciferase, and light is produced. The luminescence is proportional to the environmental concentration of the pollutant (to which the fish had been exposed), which drives the expression of the LUC gene by means of the various DNA motifs. The luminescence is quantitated in the luminometer. In each response element-containing construct, a specific class of polluting chemicals, allowing for differential identification of pollutants in a complex mixture activates the expression of the LUC gene. This assay does not require killing the fish and allows for repeated analysis of the same site with the same fish. The sensitivity of the system can be manipulated by varying the sequence of the response element.

    • PURPOSE: A measurement tub decoupling-type ecotoxicology detecting device using sulfation microorganisms is provided to protect measurement value of conductivity and pH from air and bubbles generated when water sample is injected and to prevent interruption of sulfation microorganism activities due to pH of the water samples. CONSTITUTION: A measurement tub decoupling-type ecotoxicology detecting device(10) using sulfation microorganisms comprises an inlet port(120), an outlet port(140), a bioreactor which sulfated salt ionizes oxygen by the sulfation microorganisms, a measurement tub(310) in which water sample is inflowed by being connected with the microorganism reaction tub, pH measurement unit(320) and EC measurement unit(330) which respectively measure pH and conductivity of the water samples, and a controller which judges the toxicity of the water samples by comparing the conductivity and pH with the reference. A connection pipe is respectively connected to the outlet port of the bioreactor and inlet port of the measurement tub. The inlet port of the measurement tub is placed lower than the outlet port of the bioreactor so that the water sample can be inflowed into the measurement tub. A first barrier(380) in which one side is opened is perpendicularly installed inside the measurement tub and prevents generation of turnaround flow and vortex of the water sample.

    • PURPOSE: Provided is a novel microorganism strain which has detectability for very low level of toxic substances. And a method of detecting the toxic substances using the same and a kit therefor are provided. CONSTITUTION: The novel microorganism strain, YH9-RC (KCTC 0730BP), changes the luminous level of light by reacting with toxic substances. The method of detecting the toxic substances is characterized by using aldehyde substrate to give luminescence to the microorganism. The aldehyde level used in the detection method is 0.01 to 0.015 weight. The toxic substances are selected from the group consisting of phenol, Al, As, Cd, Co, Cr(VI), Fe, Hg, Mn, Se, Zn and their mixture. The microorganism is prepared by the steps of: isolating gram negative strains from clean underground water on R2A agar plate; selecting Janthinobacterium lividium YH9 having highest sensitivity against phenol; and then introducing a luminous gene into the YH9 strain. Plasmid DNA comprising lux gene(pUTluxAB) is used to introduce the luminous gene into the YH9 strain, and the plasmid DNA is introduced into a receptor cell via a triparental conjugation.
