    • PURPOSE: The reaction of an N-(substituted methylene) glycine ester with acrylonitrile is, when necessary, followed by hydrolysis to produce the titled compound used as a synthetic intermediate of aminoacids through the shortened process in high yield.
      CONSTITUTION: A compound of formula I (R
      1 is H, methyl, aryl; R
      2 is 1W4C alkyl) reacts with acrylonitrile to give a compound of formula II. The reaction is preferably conducted in a binary-phase solvent system in the presence of a base and a phase transition catalyst. Then, the product is hydrolyzed in the presence of an acid to prepare 2-amino-4-cyanobutyric acid of formula III. The aldehyde or ketone formed as a by-product can be recycled to prepare the starting material. The compound of formula II is hydrolyzed in the presence of an alkali to produce 2-(substituted methylene)-4-cyanobutyric acid of formula IV. Compounds of formula IIWIV are useful, e.g., as a synthetic intermediate of ornithine.
